Objectives: The purpose of the study was to observe the effects of Lan Yin Feng Yang Hua Zhuo Jie Du Decoction on ulcerative colitis model rats established by trini-trobenzenesul-fonic acid/ethanol(TNBS/ethanol) through observating the general condition, disease activity index(DAI) scores and the expression of peroxisome proliferators-activated receptor-γ(PPAR-γ) 、claudin-1 and mucoprotein2(MUC2), try to explore the mechanism of the Lan Yin Feng Yang Hua Zhuo Jie Du Decoction. In order to provide experimental basis for clinical and seek new treatment methods and effective drugs.Methods: 60 male Wistar rats were divided into six groups randomly, each group 10 rats. They were marked for the normal group(NG), the model group(MG), the high dose group(HDG), the moderate dose group(MDG), the low dose group(LDG), the SASP group(SASP). All groups except NG were controlled in order to make UC animal models by TNBS/ethanol. After 3 days model-ing, the NG continued to feed, the MG were given normal saline, the other groups received liquid gavage once a day lasted 14 days. Then the stool consistency, body weight and other general condition, the colonic mucosa general concept, histopathological changes were observated. DAI scores of each group were recorded and calculated. Detected the expression of PPAR-γ、claudin-1 and MUC2 by PT-PCR and analyse effects of Lan Yin Feng Yang Hua Zhuo Jie Du Decoction on the expression of PPAR-γ、claudin-1 and MUC2.Results:1 The general case and DAI scoresCompared with the NG, the general condition of rats became lacking of energy, eating less, activity decreased, reaction reduced, weight loss, hair loose and dull, diarrhea or bloody mucus. After the 14 days treatment, There were no significant difference between the general condition of MDG and SASP, both lower than the HDG. The HDG rats were close to the NG. LDG were better than MG, and was inferior to MG group and SASP.The DAI scores:The DAI scores: Compared with the MG(2.46±0.43), the HDG(0.37±0.35), the MDG(0.80±0.28), the LDG(1.56±0.41) and the SASP(0.78±0.29) statistically significant difference(P<0.05); The HDG compared the SASP 、 MDG 、 LDG, there was statistically significant difference(P<0.05); The LDG compared the MDG 、 SASP, there was statistically significant difference(P<0.05); there was no statistically significant difference between MDG and SASP.2 The general observation and the histopathologicalThe MG was the worst. There was no significant difference between SASP and MDG, which were better than LDG while worse than HDG. The HDG was closely to the NG.The histopathologic aspects: Compared with the NG, the MG presented epithelial cells damage, a large number of inflammatory cell infiltration, congestion and edema, erosion and ulcer formation was serious. The MDG and the SASP mucosa recovered slightly worse than the HDG, but better than the LDG. The SASP mucosa recovery were close to the NG.3 The expression of PPAR-γ,claudin-1,MUC2 of ratsCompared with the NG(0.23±0.04), the expression of PPAR-γ in NG were significantly decreased, and there was statistically significant difference between them(P<0.05). Compared with the MG(0.19±0.02), the expression of PPAR-γ in each treatment group were rised obviously, there was statistically significant difference between them(P<0.05); The HDG(0.33±0.02) compared with the MDG(0.29±0.04), LDG(0.25±0.02)and SASP(0.30±0.03), Significant difference can be observed(P<0.05); The MDG compared with the SASP, no statistically significant difference( P>0.05); The SASP compared with the LDG, there was statistically significant difference(P<0.05).Compared with the NG(0.22±0.03),the expression of claudin-1 in NG were significantly decreased, and there was statistically significant difference between them(P<0.05). Compared with the MG(0.19±0.03), the expression of claudin-1 in each treatment group were rised obviously, there was statistically significant difference between them(P<0.05); The HDG(0.35±0.04) compared with the MDG(0.31±0.03), LDG(0.27±0.03)and SASP(0.32±0.04), Significant difference can be observed(P<0.05); The MDG compared with the SASP, no statistically significant difference( P>0.05); The SASP compared with the LDG, there was statistically significant difference(P<0.05).Compared with the NG(0.23±0.02), the expression of MUC2 in NG were significantly decreased, and there was statistically significant difference between them(P<0.05). Compared with the MG(0.19±0.02), the expression of MUC2 in each treatment group were rised obviously, there was statistically significant difference between them(P<0.05); The HDG(0.41±0.03) compared with the MDG(0.33±0.03), LDG(0.28±0.04)and SASP(0.36±0.06), Significant difference can be observed(P<0.05); The MDG compared with the SASP, no statistically significant difference(P>0.05); The SASP compared with the LDG, there was statistically significant difference(P<0.05).Conclusion:1 Lan Yin Feng Yang Hua Zhuo Jie Du Decoction can improve the general situation of UC rats, also have effect on the alleviation of pathological damages in rats model, and promote treatment of the ulcer. The efficacy showed a certain amount-effect relationship between efficacy and medicine dose.2 The mechanism of Lanzhusicaohuazhuojiedu Decoction on the treatment of UC may be by up-regulating PPAR-γ、clandin-1、MUC2 m RNA expression, activating gene transcription of anti-inflammatory medium, maintaining intestine mucosal barrier function, repair intestine mucosal, and promote treatment of the ulcer.
