Objective To study the effects and pharmacological mechanisms of Tuo Min Zu Fang (TMZF) in treating experimental hypersensitivity type â…  and type â…£ in vivo and in vitro. To supply some pharmacological basis for the application of TMZF in treating allergic diseases clinically.Methods (1) In vitro experiment: 1. the effects of Tuo Min Zu Fang to ileum excited by histamine. We connected the ileum of guinea pig to instrument. Experiments were composed by five parts. â‘  the effect of Tuo Min Zu Fang to normal ileum. â‘¡ Movement of ileum excited by histamine â‘¢ the treatment group of TMZF in two concentrations:1.4g/ml, 2.8g/ml and the treatment group of of chlorpheniramine (4mg/kg) . We recorded the ileum' s movement after adding different drugs. We evaluated the effects of Tuo Min Zu Fang according to the numerical values of ileum's contraction. 2. We observed the morphologic changes of Rat peritoneal mast cells which were allergized by compound48/80 to study the effects of TMZF in stabilizing the membranes of mast cell in vitro. Rat peritoneal mast cells(RPMC) were isolated through centrifugation on density gradients of Percoll. To examine TMZF' effects of stabilizing the membranes of mast cells, inhibiting degranulation of mast cells and decreasing the setting off of mediators of inflammation from mast cells, we did the following experiments: â‘ Toluidine blue was used to stain the cell models to observe the morphologic changes after treated with TMZF. â‘¡ The influence of TMZF on histamine release from RPMC elicited by compound48/80 were examined by fluorescence spectrophotometer. â‘¢ Immunohistochemistry was used to stain 5-HT in the cell models of hypersensitivity type â… . (2) In vivo experiments:We study the effects and pharmacological mechanisms of TMZF in treating experimental hypersensitivity type â…£.1. The influence of Tuo Min Zu Fang to contact dermatitis induced by cobalt chloride. Mice were allergized by PC in abdominal part, five days later, we used PC to attack the same mice' s ear. By measuring the different thickness of the mouse' s two ears, we can draw a conclusion about TMZF' s effect in treating hypersensitivity type IV.2. We also study the effects of TMZF to swelling digiti pedis induced by sheep red blood cell.Results (1) TMZF 1.4g/ml and 2. 8g/ml significantly inhibit the ileum' s movement as compared with that in the control group (p<0.01). The inhibition ratio of TMZF were 81. 39% and 92. 34% respectively. (2) TMZF 1. 4g/ml and 2. 8g/ml significantly inhibit the degranulation of RMPC as compared with that in the control group (p<0. 01). The inhibition ratio of TMZF were 73.61% and 81.73% respectively in vitro. The direct release of histamine from RPMC sensitized by compound48/80 in vitro was inhibited significantly (p<0.01) as compared with that in the control group. The inhibition ratio of TMZF were 63. 2% and 81. 2% respectively.. The contents of 5-HT in TMZF 1. 4g/ml and 2. 8g/ml was abundant as compared with that in the control group. (3) TMZF can significantly inhibit delayed allergy. In experiment about contact dermatitis, the inhibition ratio of TMZF were 54.25% and 73.86% respectively(p<0.01). In another experiment , the inhibition ratio of TMZF were 53. 07% and 78. 37%, the results were distinguished compared with that in the control group(p<0. 01).Conclusions (l)It is suggested that TMZF possesses an antagonistic effect onhypersensitivity type I and type IV. (2) TMZF may stabilize mast cell membrane andinhibit the degranulation of mast cells, and this is one of the main mechanisms of TMZF' effects of anti-hypersensitivity type I .
