Experimental Research On Gubenfangchuan Capusules To The Apoptosis Of The Asthmatic Airdrain Inflammatory Cells

[Objective]:to research the effect of the gubenfangchuan capsules on the apoptosis of the airdrain inflammatory cells of the asthmatic model and the molecular mechanism.it can provide theoretical base of clinical application and control of the asthmatic by traditional Chinese medicine.[Methods] 50 female guinea pigs of weight 200-250g were randomly divided into the normal control group, the model group ,the control group with western medicine group(dexamethasone),the low dose Chinese drugs treated group,the high dose Chinese drugs treated group, lOguinea pigs for each group. The model group ,the control group with western medicine group(dexamethasone) build the asthmatic guinea pigs model with ovalbumin derivation. The normal control group used normal saline intraperitoneal injection and atomizing . after the model was made successfully,normal saline for the normal control group and the model group dexamethasone acetate tablets, concentrated liquid of gubenfangchuan capsules (xiongfengyong xianlingpi buguzhi danshen bajitian huangqi taizishen tusizi fupian etc.)2.5g/kg and 7.5 g/kg for other 3 groups were treated 10 days .after treatment, animals were killed then we collected bronchoaveolar lavage fluid and lung tissue sample of all groups,observed the changes of LC and bronchoalveolar lavage and the changes of guinea pigs' lung tissue. Tunel France detection the apoptosis of lungs organizations eosinophil and immune organizations chemical methods observed organizations Bcl-2 and Bax protein expression of the Group guinea pig lungs . Results using statistical analysis software SPSS10.0 data processing;Count information to x+s expressed by t test comparison.[Results]: The weight of the normal control group crease naturally,without died.A guinea pig in the asthma model group died because of the asthma.The used medicine group of all guinea pigs is better than the asthma group, but it cann't return the level of normal control group, large doses concentrated liquid of gubenfangchuan can significantly reduce the apoptosis of lungs organizations eosinophil and lymphocyte of guinea pig airdrain dealing with asthma models more significant difference(P<0.01) , not significant difference with the western medicine group(P>0.05). the apoptosis of asthmatic guinea pigs eosinophil increased significantly compared with asthma model group P<0.01, not significant difference with the western medicine group(P>0.05). and increased asthmatic guinea pigs airdrain Bax expression, interferenced lungs organizations Bcl-2/Bax balance. [Conclusion]: gubenfangchuan capsule can induce or promote the apoptosis of asthmatic guinea pig EOS to alleviate the airdrain inflammatory cells exposed to control airdrain chronic inflammation, Combated asthma to the role, and through change Bcl-2 and Bax protein in asthmatic guinea pig lungs organizations expressed that the interference lungs organizations Bcl-2/Bax balance to induce the apoptosis of eosinophil and lymphocyte, then eosinophil and lymphocyte of Balf and lung...
