Objective To study the expression of PTEN protein in HepG2 cell with BMP2. To investigate the influence of BMP2 infection on PTEN expression. The relationship between expression of PTEN protein and the BMP2 was analyzed. To investigate whether BMP2 can reduce the degeneration of PTEN protein. And new pathway of HCC was studied.Methods The object was divided into three groups :control , 100ng/ml , 300ng/ml.The time was divided into there levels :6 o'clock ,12 o'clock, 24 o'clock. 10mg/ml cycloheximide were added 1h prior to BMP2 stimulation in order to inhibit PTEN expression in molecular levels .Immunohistochemical technique was used to detect the expression of PTEN in different time and different concentration in HepG2 cell. The activity of cell was detected with MTT method after interference by different concentration of BMP2 in different time :6h,12h,24h.To investigate inhibition of HepG2 cell.Result (1)The immunohistochemical result show: a,the expression of PTEN protein has significant difference in the different concentration of BMP2 groups in the HepG2 cell. There was significant difference in the positive expression of PTEN protein either between 300ng/ml groups and 100ng/ml groups and control groups (p<0.05) .b,the same result exists the different time groups. There was significant difference in the positive expression of PTEN protein either between 6h groups and 12h groups and 24 groups (p<0.05). c,The positive expression of PTEN protein was the highest in 300ng/ml and 24h groups. There are not interaction between density and time. (2) MTT result show: The growth of HepG2 cell was inhibited at any time (6h,12h,24h).The inhibition of HepG2 cell growth was the highest in 24h groups. There has relationship between the inhibition of HepG2 cell growth and the concentration of BMP2. The inhibition of HepG2 cell growth was the highest in 300ng/ml groups. There was significant difference in the inhibition of HepG2 cell growth in different time and different concentration groups(p<0.05).Conclusion (1)BMP2 can increase the expression of PTEN protein in HCC. There has dependence of dose between BMP2 concentration and time and the expression of PTEN protein. BMP2 influence on PTEN protein by reducing degeneration of PTEN protein mainly. (2) The increase of PTEN protein can inhibit growth of cells in HCC.
