Objective:To observe the expression of P53and PTEN in urothelial carcinoma and normal mucosa of bladder, and to expound the potential relationship between the expression of P53,PTEN and clinicopathologic factors such as age, gender, smoking history, incipient or recurrent,tumor stage and histologic grade of bladder urothelial carcinoma. To discuss the correlation between the expression of P53,PTEN in Bladder transitional cell carcinoma(BTCC).Method:We collected68cases of bladder urothelial cell carcinoma and25cases of benign prostatic hyperplasia which were all confirmed by pathology specimens in August2009-August2011. BTCC patients’average age is63with a range of42-98years. The result were observed by light microscope, and buffy particles in cells were considered as positive cases. The expression of P53,PTEN in11normal bladder mucosa and95bladder BTCC tissues were detected using immunohistochemical study, and correlated with tumor stage, histologic grade, and patients’ age, gender, smoking history. Data were performed with SPSS17.0software. Chi-square test and Pearson rank correlation were used. Data were considered statistically significant when p values were less than0.05.Results:In68cases of BTCC,45patients showed positive expression of P53, the positive rate was66.18%; In the11cases of benign bladder tissues, the expression were2cases, the positive rate was18.18%. Two groups were compared. The difference of BTCC and benign benign bladder tissues group was statistically significant(P<0.01). In68cases of BTCC,23patients showed positive expression of PTEN, the positive rate was33.82%; In the11cases of benign bladder tissues, the expression were10cases, the positive rate was18.18%. Two groups were compared. The difference of BTCC and benign bladder tissues group was statistically significant(P<0.01).Expression of P53was not correlated with expression of PTEN(P>0.05).Conclusion:1. The expression of P53in BTCC tissues was significantly higher than in normal bladder mucosa, The expression of PTEN in BTCC tissues was significantly lower than in normal bladder mucosa,which reveals that P53,PTENmay play an important role in the occurrence and progression of BTCC.2. The expression of p53,PTEN in bladder urothelial carcinoma tissues was positively correlated with tumor clinical stage, pathological grade; Compared with incipient cases, there was not a higher positive rate of the th two proteins in the recurrent cases. To detect the expression of P53,PTEN contributes to diagnosis and prognosis of BTCC.3. Expression of P53was not correlated with expression of PTEN(P>0.05).The expression of P53,PTEN become a new target for targeted therapy of BTCC.4.The combined detection of P53,PTEN proteins can be used to evaluate the incidence of BTCC development, prognosis, and provide an important basis for drug treatment.
