Experimental Study On Effection Of The Soluble Proteome In HepA Carcinoma Of GAPSGF Injection

Objective:To study the effect of GASPGF on the soluble proteome of HepA carcinoma,we utilized the proteomics technology to further study GASPGF anti-tumor function mechanism.Methods(1)The ratio of antitumor:weigh the intact tumor of each group and calculate the ratio. (2)Using SDS-PAGE to separate the protein ,observe the effect of GASPGF on soluble proteome of HepA carcinoma;(3)Using two dimensional ployacrylamide gel electrophoresis(2-DE) to separate the protein,observe the effect of GAPSGF on soluble proteome of HepA carcinoma.ResuIts:(1)Campared to tumor group,GASPGF group have notable difference on the ratio on antitumor (P< 0.05);(2)In 1D-SDS-PAGE of soluble proteins, 16 bands were showed in control group,24 bands in tumor group and 18 bands in GAPS group. Compared with control group, 3 bands were down-regulated and 11 bands were up- regulated in tumor group. GAPSGF could make 3 down-regulated bands and 8 up- regulated bands come back.(3)In 2-DE of soluble proteins, 101 spots were showed in tumor group and 85 spots in GAPS group. 54 spots only showed in tumor group and 38 spots only showed in GAPS group. There were 14 spots which showed both in tumor group and GAPS group but the quantity were different more than 2 times.Conclusion: (1)GAPSGF has obvious antitumour effect on HepA carcinoma. (2)GAPSGF can effect the expression of soluble proteins in HepA carcinoma(1)There are 54 spots only expressed and 6 spots expressed decreasing in tumor group,these proteins maybe were expressed by oncogene about the carcinomar. (2) There are 38 spots only expressed and 8 spots expressed increasing in GAPSGF group,these proteins maybe were expressed by aitioncogene about antitumor effecet of GAPSGF.(3)On proteome, GAPSGF could down-regulate the expression of oncogene and up-regulate the expression of antioncogene,the antitumor effect of GAPSGF maybe about it .
