Aching is the main clinical symptom of ulcer disease, illness position of which is stomach ,and involving in liver and spleen. The stomach is the FU-organ which has affluent qi and blood, the liver stores blood, and the spleen control blood to avoid bleeding, under pathologic state, various factors can result in the liver failing to maintain the normal flow of qi, the ascending and descending of qi activities of spleen and stomach abnormally which leading to the malfunction of the flow of qi and blood , the stagnation of blood in stomach of collaterals and subcollateralsv. In addition, peptic ulcer with long course of disease often attacks recurrently, and it is cured difficultly. Since "pain caused by stasis", " the evil transmits to the collaterals and subcollaterals when attacking the body long", "pain long lead to stagnation of blood", and the qi deficiency and qi stasis also result in the stagnation of blood. Therefor, the stagnation of blood exist in peptic ulcer with various degree. The treatment method of promoting blood circulation to resolve stasis has the profound and far-reaching significance for preventing and treating peptic ulcer. This study aims to observe the effect of anti-ulcer of Danshen Yin based on the correlated clinical study and pharmaco-investigation to provide objective evidences from the two aspects of pharmacodynamics and effective mechanism, and to verify the effect of the treatment method of activating blood circulation to dissipate blood stasis in preventing and treating peptic ulcer.Duplicate rat peptic ulcer model by means of pyloric ligature, acetic acid daub and water immersion stress methods to observe the effects of Danshen Yin on experimental gastric ulcer andexplore the potential antiulcerative mechanism of action in preventing and treating peptic ulcer. The results of this experiment demonstrate that Danshen Yin has conspicuous effect of preventing and treating action on gastric ulcer caused by water immersion stress method and acetic acid daub method. Danshen Yin can not play roles on gastric juice quantity,total acidity and pepsin activity of gastric fluid in gastric ulcer rats due to ostium pyloricum deligation, but can increase the contents of NO in gastric fluid (compared with model group P<0.01), increase the activity of SOD in gastric tissue and decrease the contents of MDA in gastric tissue (compared with model group P<0.05). Besides, in the model of gastric ulcer caused by acetic acid daub method, Danshen Yin can increase the contents of NO in rats blood serum and the contents of 6-keto-PG in rats blood plasma(compared with model group P<0.01). These manifest that the relative mechanisms of Danshen Yin to anti-gastric ulcer may be as follows: improve the gastric mucosal defensive function by increasing the synthesis and release of NO> 6-keto-PG which were protecting factors of gastric mucosa;improve the capability of clearing away oxyradical to promote the recovery of gastric mucosa.This study provides experimental evidences for preventing and treating peptic ulcer clinically of Danshen Yin, verifies the effect of the treatment method of activating blood circulation to dissipate blood stasis in preventing and treating peptic ulcer, and possesses certain reference value for directing the application of drugs to preventing and treating peptic ulcer and developing new drugs.
