Safflower, Chinese traditional medica for invigorating the circulation of blood and reducingstasis, is the dried flower peals of Carthamus tinctorius L, in which carthamus is one of the most important components.The extraction, purification, structure, standard, determination, pharmacokinetics and the antioxidation effect of carthamin was systemically studied in the thesis. The results were as follows:(1) The extraction and purification of total carthaminwith the content of carthamin as an index , the orthogonal design of four factors and three levels were used to select the optimum extraction process. The optimum extracting condition was to add 10 times amount of water(pH=10), soaking and extracting for 20 minute every time , nine times totally. The said solution was adjust to pH=4 and the precipitate was separated from the solution.(2) The research of preparation new method, structure identification and standard for carthaminThe micrite cellulose column chromatograph and the Sephadex LH-20 column chromatograph, a new method on the preparation of carthamin was determined. This method is simple and stable. Carthamin of purity index isolated by use of the above method was up to 98.1% by HPLC , and corroborated as carthamin acorrding to physical & chemical properties and its spectrometric data(UV,IR,MS,1H-NMR,13C-NMR and DEPT). The standard of carthamin was completed.(3) Quantitative determination of carthamin by RP-HPLCThe RP-HPLC method was used to determine the content of carthamin in Carthamus tinctorius L. The analysis was carried out on Phenomenex Luna C18 column(4.6mm×150mm,5μm). The mobile phase is CH3CN-CH3OH-H2O(35:15:50, V/V). The flow-rate was 1.0mL/min and wave-length at 518nm and temperature at 25℃. The method was simple and had a good linear relationship . The linear range is 0.1441.44μg and give a correlation (r) of 0.9996. The recover was y=4.184531×106x-4858, The minimum content of carthamin should be 0.042%.
