Objective To investigate the expression and clinical significance of PTEN,VEGF and Ki-67 protein in human astrocytoma.Menthods The expressions of PTEN,VEGF and Ki-67 protein were examined by S-P immunohistochemical staining in 65 human astrocytomas and 15 cases of normal brain tissues.Results 1. PTEN was detected 49.23% in astrocytoma. The expression of PTEN trended to be lower in astrocytoma with high-grade WHO classification. The expression of PTEN were significantly different between low-grade group and high-grade group(P<0.05). No significant different expression of human astrocytoma was detected betweenâ…¢,â…£grade(P>0.05). 2. Positive rate of VEGF expression was 64.6%. No VEGF was examined in the control group. There was a significant VEGF expression between low-grade group and high-grade group (P <0.01). No significant different expression of human astrocytoma was detected betweenâ…¢,â…£grade(P> 0.05). 3. The expression of Ki-67 significantly correlated with the pathological grade of astrocytoma. 4. The expression of PTEN had a negatively correlated with VEGF, Ki-67. There was no significant correlation between VEGF and Ki-67. 5. PTEN protein did not correlate with patient gender, tumor size and tumor location. However, there was significant difference between 44-younger group and 44-year-older group (P=0.03). It showed that more inactivation of PTEN occured in the patients of elderly. 6. The expression of VEGF did not correlate with patient age, gender and tumor location, there was significant difference between 5cm-smaller group and 5cm-bigger group(P=0.01). It showed that higher expression of VEGF in human astrocytoma of large size and rich in vascular. 7. Ki-67 expression had no significant correlation with of patient gender , age, tumor location and tumor size ( P > 0.05).Conclusion The inactivation of PTEN in astrocytoma may lead to a over-expression of VEGF and Ki-67, to promote astrocytoma angiogenesis and tumor cell proliferation. The three proteins interact during tumor proliferation and invasion.
