Objective: Gliomas are the most common primary brain tumors. the prognosis of patients with malignant gliomas is very poor. With the increase of knowledge in tumor biology and molecular genetics, we know the survival rate is related with the adapt of the oxygen-poor environment,increment of cell out of control,Vascularization,extensive brain metastases and so on. angiogenesis in the tumor has become one of the foremost topics of interest in biomedical research. PTEN,Hypoxia inducible factor-1 (HIF-1) and VEGF plays a critical role in angiogenesis during vascular development in the tumors. Our study is to investigate the expression of the hypoxia-inducible factor(HIF-1α)and vascular endothelial growth factor(VEGF) in human golimas and to analyze the relation among them.Methods: From March.2004 to may.2006, we collected 62 samples of human gliomas and 6 samples of normal brain tissues from the patients,received the operation in the second hospital of hebei medical university. The study was focused on the expression of PTEN,HIF-1αand VEGF. Expression of the PTEN,HIF-1αand VEGF was investigated by immunohistochemistry in 62 specimens of human gliomas. The microvessel density (MVD) was observed by microscopy.Results: this experiment altogether examined 62 samples of human gliomas and 6 samples of normal brains, the expression of them as follow:1 6 samples of normal brains have PTEN to express. The expression levels of PTEN was decreased corresponding to the ascending of the glioma grades, the level of PTEN was 85.7 in glioma gradeⅠ-Ⅱ,63.6% in glioma gradeⅢ,50% in glioma gradeⅣ. The expression of PTEN was correlated with the glioma grades.2 6 samples of normal brains does not have HIF-1αto express. The expression levels of HIF-la was increased corresponding to the ascending of the glioma grades, the level of HIF-1αwas 39.3% in glioma gradeⅠ-Ⅱ,81.8% in glioma gradeⅢ,100% in glioma gradeⅣ. The expression of HIF-1αwas positively correlated with the glioma grades.3 6 samples of normal brains does not have VEGF to express. The expression levels of VEGF was increased corresponding to the ascending of the glioma grades, the level of VEGF was 60.7% in glioma gradeⅠ-Ⅱ,86.4% in glioma gradeⅢ,100% in glioma gradeⅣ. The expression of VEGF was positively correlated with the glioma grades. 4 The normal brain organization and the tumor organization in nature blood vessel endothelial cell hasⅧfactor related antigen expression. The level of MVD was 6.20±0.84 in normal brains. 14.12±1.57,28.53±2.20 in glioma grade(Ⅰ,Ⅱ),46.71±3.73 in glioma gradeⅢ,79.36±2.87 in glioma gradeⅣ.5 The expression levels of HIF-1αand VEGF was decreased corresponding to the expression levels of PTEN. The expression levels of VEGF was increased corresponding to the expression levels of HIF-1α.Conclusion : The expression levels of PTEN was decreased corresponding to the ascending of the glioma grades. The expression levels of PTEN,HIF-1α,VEGF and MVD was increased corresponding to the ascending of the glioma grades. The expression of HIF-1αwas also positively correlated with VEGF and MVD expression. PTEN,HIF-1αand VEGF may play a critical role in gliomas regarding their role in angiogenesis.
