Objective: To investigate the expression of isoform AML1/ETO9a in the acute myeloid leukemia (AML)- M2 patients and its clinical significance .Methods: Expression of AML1/ETO fusion gene and isoform AML1/ETO9a were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) in AML- M2 patients at preliminary diagnosis , in relapse, respectively , as well as in patients with other hematologic deseases and in healthy controls. Karyotype was studied by R-binding technique.Meanwhile,the clinical significance of isoform AML1/ETO9a was analyzed by following-up some patients from preliminary diagnosis for the CR rate and survival time,.Result: 1. Among 48 of 100 cases with t(8;21) at initial treatment , expression of AML1/ETO9a could be detected in 46 cases at first presentation and in 11 of 22 patients in relapse , while no expression of this isform could be found in the rest. 2. Expression of AML1/ETO and AML1/ETO9a could be detected in 25 of 55 cases , which was be followed-up ; only expression of AML1/ETO could be detected in 2 cases ; only expression of AML1/ETO9a could be detected in 2 cases ; no expression of either could be found in the rest . 3. 55 cases, which could be statisticsed the curative effect, were treated by combined chemotherapy. 47 cases(85.5%) achieved complete remission ; 5 of 27 patients with isoform AML1-ETO9a positive didn't achieve complete remission ; 3 of 28 patients with isoform AML1-ETO9a negative didn't achieve complete...
