The pharmacologic actions of Dahuang-Fuzi herb-couple changed with the proportion of each herb, but the chemical foundation and involved mechanisms have not been revealed. This study investigated the chemical chang according to the contents of the components in the herb-couples and the changes of alkaloids in the Fuzi and its processed Fuzi, such as Baifupian, Heishunpian and Yanfuzi.An RP-HPLC method for simultaneous determination of chrysophanol, emodin, rhein, aloe-emodin and physcion has been developed to investigate the change of chemical constituents of the herb-couples. The linear ranges of chrysophanol, emodin, rhein, aloe-emodin and physcion were 10.60-212.0 (r=0.999 5), 2.720-54.40 (r=0.999 3),2.520-50.40 (r=0.999 6),2.560-51.20 (r=0.999 7) and 2.760-55.20μg·mL-1(r=0.999 3), respectively. The average recoveries (n= 5) of the five anthraquinones were (101.1%±1.8%)、(102.8%±2.6%)、(96.7%±1.5%)、(100.9%±2.1%)、(99.6%±2.4%).The contents of the five anthraquinones in the herb-couple were changed compared with Dahuang only. The unitary diversity of the ration 1:1 were the greatest via the Minknowski.Three diterpenoid alkaloids were simultaneously determined by the method of RP-HPLC in different processed Fuzi(Baifupian heishunpian and yanfuzi). The linear ranges of aconitine、mesaconitine and hypaconitine were 1.004-40.16 (r= 1.000),10.00-350.0 (r=0.999 7),10.00-300.0 (r=0.999 5)μg·mL-1, respectively. The average recovery (n=5) of the three diterpenoid alkaloids were (96.7%±1.5%)、(101.5%±1.8%)、(100.0%±2.4%). The amounts of hypaconitine was the highest,and mesaconitine was higher than aconitine in Shengfuzi. The three alkaloids in the processed Fuzi were much more decreased compared to Shengfuzi.The three diterpenoid alkaloids were also determined in the Fuzi combined with Dahuang at different proportions. The linear ranges of aconitine mesaconitine and hypaconitine were 1.004-40.16 (r 0.999 9),10.00-300.0 (r=0.999 8), 0.01000-0.3500μg·mL-1 (r= 0.999 8), respectively. The average recoveries (n= 5) of the three alkaloids were (105.1%±2.6%)、(103.2%±1.8%)、98.3%±2.3%). The amounts of three alkaloids in the Shengfuzi were decreased after combined with Dahuang.The aconitum alkaloids from Fuzi and its processed samples were identified by using HPLC-TOFMS/MS. Both the number and the contents of alkaloids characterized in the samples of processed Fuzi were decreased.
