Objective:Research the basic toxicity of crude lateral root of aconite, black prepared lateral root of aconite and white prepared lateral root of aconite to many animal models with combination of illness and syndrome, such as discover the dependability between the decoction time, administration dosage, drug processing, animal models with combination of illness and syndrome and the toxicity of lateral root of aconite; Explore the application methods of animal models with combination of illness and syndrome in the research of Toxicology of Traditional Chinese Medicine. Methods:On the base of the clinical actions and indication of lateral root of aconite and under the specified-pathogens free (SPF) experimental animal barriered system surroundings, select the SPF experimental animal to build the animal models of pain, inflammation, syndrome of deficiency of spleen-YANG, syndrome of deficiency of kidney-YANG, and study the acute toxicity of crude lateral root of aconite, black prepared lateral root of aconite and white prepared lateral root of aconite, after decocted for 15 minutes, 30 minutes, 1 hour, 2 hours, 3 hours, 4 hours and 6 hours, to all kinds of animal models with combination of illness and syndrome; meanwhile, combined with the methods of uniform design and regression analysis, study the long-term toxicity of crude lateral root of aconite, after decocted for 15 minutes, 30 minutes, 1 hour, 2 hours, 3 hours, 4 hours and 6 hours with the administration dosages of 72 times, 12 times, 120 times, 48 times, 6 times, 96 times and 24 times of clinical dose to the animal model of asthenia cold syndrome.Results:1.The acute toxicity research results of the effect of lateral root of aconite on the animal models with combination of illness and syndrome:(1) With the crude lateral root of aconite decocted for 15 minutes to 4 hours and through the acute toxicity observation of model animals, the result is that all the model mice of pain, inflammation, syndrome of deficiency of spleen-YANG and syndrome of deficiency of kidney-Yang have the toxic symptom of hiccup, motor paralysis, salivation, diarrhea, hypnody and hyperspasmia, with the medial lethal dose (LD50) respectively...
