Objective: The experiment will study the anti-hepatic fibrosis effect and mechanism of the Astragalus Root, Chinese Angelica Root, Bupleurum Root and their combination in rats. The two objectives are as followed: (l)To investigate if the interaction exits when the Astragalus Root , Chinese Angelica Root and Bupleurum Root are applied to treat the CCl4-induced hepatic fibrosis of rats.(2)To study the best dosage ratio of the three traditional Chinese herbs in the treatment of experimental hepatic fibrosis model of rats in order to offer experimental reference to the clinical application.Methods: The experiment is composed of two parts.The experimental design adopts factorial design and orthogonal design with three factors and two levels.In the experiment, we choose 72 healthy male rats to divide into 18 groups by randomization, 4 rats in each group(including,each 8 groups in Part A and Part B, each 1 group in normal group and model group) The drugs are used at the beginning of the making model.The rats will give the combination of the three drugs with different dosages.The two levels of part A are having and not having drugs. The two levels of part B are high and low dosages.The total treatment period is 6 weeks. At the end of treatment ,all of the rats would be killed to get the serum sample to test the serum IV-C, HA by radioimmunossay and the MOD value of immunohistochemistry staining of LN and α-SMA.At last,the liver tissue is taken to make the HE staining ,VG staining and take photos. Results: (l)The three traditional Chinese herbs and their combination have significant anti-hepatic fibrosis function.The interaction exits mainly among Astragalus Root and Chinese Angelica Root, while they are applied to treat the experimental hepatic fibrosis of rats. (2)The Dose-Reaction Relation among the three herbs in the experiment is different because of different hepatic fibrosis index.Conclusions: The Astragalus Root, Chinese Angelica Root, Bupleurum Root and their combination have the anti-hepatic fibrosis effect on rats at various degree. The interaction exits when they are used together beause the anti-hepatic fibrosis mechanism of three herbs are different. Therefore, it is necessary to study the best Dose-Reaction Relation of the three herbs in the prescription.
