This thesis establish a HPLC method for the determination ofcontent of chuanlong guci Capsule. Method: Determine the content ofchuanlong guci Capsule by HPLC. The condition of chromatogram and systemsuit test: with C18 column(4.6×150mm, 5μ),the mobile phase wasacetonitrile-water(82∶18) at the flow rate 1mL·min-1, with UV detection atwas 203nm.The column temperature was 35℃, Plates shouldn't be under4000 according to Diosgenin.Preparation of contrast solution: Take the right amount the contrast ofdiosgenin ,accurately weight, put methanol in it to preparate the solution of0.2mg·ml-1,it's all right. Preparation of sample contrast solution: Mix anamount of the capsules contents, equivalent to about 1g , accurately weight,with about conical flask ,put 50.0mL methanol in , accurately weight,withultrasonic wave 50 minutes, and cool ,accurately weight,supplement the lost.take filtrate 25.0mL to conical flask ,vaporize, solution with 3mol·L-1 HCL inwater bath 30 minutes, put methylene choride 30mL circumfluence, putmethylene choride 30mL the same way , vaporize, solution and transfer to10-mL volumetric flask. it's all right.The method of determine:Respectively accurately draw contrastsolution 10μL and sample solution 10μL, inject HPLC instrument, determine,it's all right. Results: Liner test: The regression equation is Y= 757.5X-5.4508r=0.9999(n=5),as a result declare: Diosgenin showed a good linearrelationship at a range of 0.64μg-3.2μg.Accuration test: with RSD of 0.17%,as a result declare: The instrument possess better accuration. Reappear test:with RSD of 0.79%, as a result declare: The method's reappear is good.Stable test: with RSD of 0.76%, as a result declare: the properties ofdiosgenin is stable in twenty four hours. Recovery test: The averagerecovery is 99.2%, RSD=0.3%,(n=6).Conclusion: The method is simple, rapid, reliable, It can be used forthe quality control of chuanlong guci Capsule.The thesis has studied the stability of chuanlong guci Capsule to findout that their quality is stable according to the property and content ofchuanlong guci Capsule placed nine months.
