Study On The Anti-breast Cancer Effect And Mechanism Of Bidirectional Fermented Extract Of Huaier Banlangen Root

Bidrectional fermentation is called the medicianal fungal new type(bidirectioanl)solid-state fermentation engineering,that the medicinal fungi is used as fermentation strain,the active ingredients of Chinese herbal medicines as medicinal substance to replace the traditional fungal culture(bagasse,wheat bran,and other agricultural products).The potency substance in the fermentation process provides nutrients such as carbonitride which fungal requires,the enzyme which is produced by the fungus can change the component of herbs,achieve better efficacy,and reduce the toxicity of raw herbs.Bidirectional fermentation technology can be applied without separating with medicinal fungal's secondary metabolites separation and work together to make better medicinal effect.Breast cancer is one of the malignant tumors that are harmful to women's health.In recent years,the incidence of breast cancer is rising.At present,the most effective treatment for breast cancer is surgical treatment,preoperative or postoperative adjuvant radiotherapy,chemotherapy or immunotherapy.Many chemotherapeutic agents can effectively kill tumor cells,but their toxic side effects and many drug resistance limit the clinical applications.Therefore,how to solve the problem of clinical treatment of breast cancer,and related basic research.People began to attach great importance to these issues.This study is on the basis of the previous research of our research group.Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation productanti breast cancer effects and the mechanism of action for more in-depth research.The pharmacodynamics as the evaluation index,not only for determine Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product whether anti breast cancer and whether separate Trametes robiniophila Murr and Radix Isatidis has more advantages to provide scientific data,but also for the compatibility of traditional Chinese medicine theory and modern processing methods as bio fermentation technology of science to provide more data.The methods and results of this study are as follows:(1)Trametes robiniophila Murr is inoculated in Radix Isatidls herb.The quality of Trametes robiniophila Murr and Radix Isatidis extraction with 70%ethanol,have Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation extract product.The breast cancer cells with different expression types of estrogen were expressed in 3 different lines.The effects of Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product on proliferation and apoptosis of breast cancer cells.The results showed that Trametes robiniophila Murr growth in good condition;Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product could significantly inhibit cell proliferation,and there is a dose and time effect relationship.Compared with Radix Isatidis and Trametes robiniophila Murr,Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product were better inhibitory effect on cell.Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product can block the cell cycle,induce apoptosis of 3 line breast cancer cells.(2)The effects of Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation producton metastasis and invasion of breast cancer cells.Cells scratch assay,Transwell assay and cell adhesion assay results show that the Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product can significantly inhibited cell healing,the number of cells through the membrane matrix and cell adherent ability.Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product inhibit breast cancer cell migration and invasion ability better thanTrametes robiniophila Murr and Radix Isatidis.The compatibility of Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product has better anti tumor cell invasion and metastasis effect shows.Trametes robiniophila Murr and Isatidis Radix bidirectional fermentation products may down-regulate the expression of MMP-9 to inhibit the migration,invasion and adhesion capabilities of breast cancer cells.(3)Study on the inhibitory effect of Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product in 4T1 mousebreast cancer engrafted tumor.Results show:Trametes robiniophila Murr and Radix Isatidis bidirectional fermentation product can inhibit tumor growth and has a protective effect on immune system in mice.
