Objective To address paraoxonase 1(PON1) gene polymorphism and its relationship with plasma paraoxonase activity , lipid and lipoprotein levels in preeclampsia. Investigate expression of PON 1 and PON2 in placenta.Methods Lipid and lipoprotein were detected with automatic biochemistry analyzer. Paraoxonase activity was measured spectrophotometrically with phenylacetate. Gene polymorphism of paraoxonase at site 192 was detected with polymerase chain reaction(PCR)and restriction fragment length polymorphism(RFLP).Results(1) In severe preeclampsia group, the levels of total cholesterol(TC),triglyeride(TG),low density lipoprotein(LDL) and apolipoprotein B(apoB) were higher than that of health gravida group(p<0.05),while high density lipoprotein and apolipoprotein A (apoA) were lower. The levels of TG,HDL,apoB and apoA were significantly different between mild preeclampsia and health gravida. The level of CHOL and LDL were significantly different between the mild preeclampsia group and severe preeclampsia group(p<0.05), while there were no significant differences between mild preeclampsia group and health gravida group(p>0.05).(2) The PON1 activity was lower in preeclampsia group than that in health gravida group(t=16.11,p<0.05).No significant difference was found between severe preeclampsia and mild preeclampsia(p>0.05).(3) There was correlation between PON1 and apoA,HDL and LDL, and the respective... |
