Objective: Through studying the changes of the contents H2O in spinal cord tissues after ASCI, observing the changes histopathololgy, to investigate the possible mechanism on protective effects of vitamin E on the injured spinal cord in rabbits so as to seek a new way to alleviate the secondary spinal cord injury.Methods: Fifty one New Zealand white rabbits were randomly assigned to three groups: The control group (Group A n=17); The acute spinal cord injury (ASCI) group (Group B n=17); The ASCI with vitamin E treatment group (Group C n=17). The rabbits were injected 3% nembutal (1mg/kg) intraperitoneally. Secondly, cut L1-3 spinous process and lamina of vertebra to expose dura. Group A is laminectomy-only and no trauma. The ASCI animals were made by using an improved Allen's weight-drop device. 270g·cm contusion injury was introduced to Group B and Group C. After thirty minutes the animals of Group B were injected 1ml/kg physiologic saline through intraperitoneum and Group C were injected 10mg/kg vitamin E. Ten rabbits were chose to kill from every group randomly after 48 hours. Spinal cord samples were obtained from original wound center. Every sample is 0.8cm. The samples were divided into two parts from center. One half was placed in 10% formalin. After 24 hours they were taken out to make the dyeing of HE and observed the histopathology through light microscope. Another half were used to examined the contents of H2O. Two rabbits of every group were perfused and taken the spinal cord samples to make TEM slices were observed and taken photos through H-7500 TEM. Inclined plate+Gale scale were observed 2-week after ASCI with the five-remainder rabbits of every group. The result was analyzed through SPSS 12.0 software. Analysis of variance(ANOVA) and t-test were used for the contents of H2O and inclined plate scale.
