Objectives'. (1) Study the procedure of extracting evening primrose oil (EPO) with supercritial CO2 fluid;(2) Evaluate advantages of extraction- fractionation using supercritical CO2 to extract EPO;(3) Establish procedures of determining the content of γ-linolenic acid in EPO, and preliminary techniques of evaluating the quality index of the EPO for injection;(4) Research techniques on the preparation of EPO for injection use. Methods: (1) Establish the methodology of determining the content of γ-linolenic acid in EPO with the gas chromatography internal standard method and preliminary analysis methods on evaluating the quality index of the EPO for injection by referring to the "National Pharmaceuticals Standards" and "Chinese Pharmacopoeia" (2005, Part II) - soybean oil (for injection);(2) According to the oil-extracting rate, the content of γ-linolenic acid in the extraction product and the EPO quality index, study EPO extraction conditions with supercritial CO2 fluid and separation conditions of the fractionation column;(3) According to the "National Pharmaceuticals Standards" of EPO, compare the quality difference of EPO prepared by (a) the extraction-fractionation technique of using supercritial CO2 fluid, (b) regular supercritial CO2 fluid extraction technique and (c) organic solvent extraction technique;...
