Studies On Main Components Of Evening Primrose Seed

In order to find out the pharmaceutical bioactive-components from the seeds of Evening Primrose comprehensively and accurately,and promote the exploitation and utilization of its whole resource,main components from seeds of Evening Primrose were extracted,isolated,purified and identified in turns.The results of pre-testing indicate that there are alkaloid,amino acid, protein,reducing sugar,stearic acid and coumarin in the seeds,and maybe presents phenol,saponin,triterpene and sterol,but no tannin and cardiac glycoside present.The seeds were first crushed and extracted with 75%ethanol,the extract was condensed.The condensate was extracted with petroleum ether,ethyl acetate,n-butyl alcohol and methanol in turn.The extracts were analyzed by thin layer chromatography(TLC)to ascertain the suitable solvent elution systems for each extractive.Then the extracts were further isolated with silica gel column chromatography and HPLC.We investigated the optimum conditions for each extractive with HPLC and prepared the purified products and obtained three compounds.These three compounds were analyzed and identified by IR,NMR,MS.Two compounds were identified to be substance with a glucose-glucose-rhamnose main-chain and it should be an oligo-saccharide and molecular formulas of C₂₀H₃₅Cl.The third compound was identified to have a glucose-rhamnose main-chain with molecular formula of C₁₅H₂₆O₁₀.In addition,we studied the elimination on H₂O₂ of crude extract from the seeds.The results indicated the aqueous extract and ethanol extract both exhibited certain limitation ability,thus crude extract had definitive ability on the elimination of free radical and retardance of lipid peroxidation,it had special meaning in human anti-disease,body health and ageing-prevention.The study will provide theoretical basis for comprehensive utilization of Evening Primrose and promote the exploitation of Evening Primrose.
