Objective: Detecting the histamine release ratio from in vitro sensitized normal mast cell ,discuss the antibody type in guinea pig allergy; identify the main allergen of some normal food and analysis the cross reactivity.Methods: The guinea pigs were immunized with extract of shrimp protein, cashew protein and soybean protein with the adjuvant of aluminum hydroxide. The specific IgE and IgG of food allergens was measured by indirect ELISA. IgG1,IgE and the mixture of other subclass of IgG of the immune serum of sea shrimp allergic models were purified by the protein A. The mast cells of normal guinea pig were collected, stimulated by the antibody IgE, IgG1 which purified from the immune serum in vitro. The histamine release ratio were detected by HPLC method. The main allergen components in food allergen extract were identified by Western blot, and purified by anion exchange chromatography. The cross activities between different food were analyzed by Western blot.Results: The levels of slgE in allergic guinea pig model were higher than those in control. It proved that the allergic models established successfully. The slgE was the highest at 14th day after challenge, reached 1/80, 1/40 and 1/80 in shrimp, soybean and cashew group respectively. The result of histamine release ratio of the normal mast cell stimulated by total serunu IgE and IgG is 52.46±2.47% ,40.05±1.98% and 71.43±3.47%.This result showed that IgG and IgE are all taking part in occurring of guinea pig allergy. In four types of IgG subclassjgd is the main part and the other subclass didn't take part in. The main allergen component of shrimp , which molecular weight is 21,36,38 and 40kD,and have cross activity with oyster. the main allergen component of cashew, which molecular weight is 55kD,28kD 和 14kD, the main allergen component of soybean is 14,28,30 and 60kD, had been identified by Western blot analysis.Conclusion: Through establishment of guinea pig allergy model, the main antibody and subclass of guinea pig allergy have been preliminary identified.The commen food , shrimp and oyster .cashew and soybean have cross allergen components.The detecting of histamine release of mast cell stimulated in vitro possible be a new way of analysising and identifying allergen.
