Swertia macrosperma C. B. Clark grows widely in Tibet,Guizhou and Yunnan.It is a kind of Tibetan drug with wide treatment functions. We studied the chemical constitunts of Swertia macrosperma C. B. Clark from Tibet, determinated the effective constituents in different parts by HPLC, and explored the antioxidative effect of the crude extracts and several pure compounds.Ten compounds have been isolated from Swertia macrosperma by kinds of column chromatography, such as silica gelcolumn chromatography, thin layer chromatography and recrystallization. seven of the ten compounds have been identified as 1 , 8-dihydroxy1-3 , 5- dimethoxyxanthone(swerchirin , I) , 1-hydroxy1-3, 5-dimethoxyxanthone (II), 1, 8-dihydroxyl-3, 7-dimethoxyxanthone (methylswertianin, III),1-hydroxy1-3, 7, 8-trimeth-oxyxanthone(decussatin, IV), β-sitosterol(VI),oleanolic acid(VIII) and 1, 5, 8-trihydroxy1-3-methoxyxanthone (bellidifodin, IX) respectively by UV, IR, 1HNMR, 13CNMR and MS analysis. Compound I~VI were obtained from this plant for the first time.Established a method for the content determination of decussatin, bellidifodin, methylswertianin swerchirin and oleanolic acid in Swertia macrosperma C. B. Clark. The calibration curves of four xanthones and oleanolic acid were in good linearity over the range of 0.154~0.77 μg, 0.144~0.72 μg, 0.172~0.86μg, 0.224~1.344μg 和 2.112~10.56 μg, respectively (r=0.9999), the average recoveries were 98.18% (RSD= 1.19%), 99.32%(RSD=1.52%), 98.44% (RSD=1.25%), 97.68% (RSD=1.57%) and 97.18%(RSD=1.32%), respectively. The method is rapid and precise.The antioxidative effect of Swertia macrosperma was studied.
