Objective:To compare the effects of three methods on the rabbit model of traumatic proliferative vitreoretinopathy.Methods:Twenty-four pigment rabbits,48 eyes in total, were randomly divided into four groups of 12 eyes each on the basis of all causing rabbit eye trauma, three experimental groups:Group A:0.3 m L platelet-rich plasma(PRP),Group B:0.3 m L transforming growth factorβ2(TGF-β2)(50 ng/m L),Group C:0.3 m L retinal pigment epithelium(RPE)(1.5×10 6/m L),control group:Group D:0.3m L saline.Postoperatively, the proliferation of vitreous and retina was observed weekly by fundus photography and ultrasound;the expression ofα-smooth muscle actin(α-SMA)during intravitreal retinal proliferation was detected by western blot(WB)at the fourth week postoperatively and the retinal tissue structure of each group was observed under a microscope by frozen section. Results:At the fourth week after surgery, the mean grades of traumatic proliferative vitreoretinal lesions in the control group and the three experimental groups were 0,3.67,2.33,and 2.50,respectively;the grades of traumatic proliferative vitreoretinal lesions groups were significantly higher in the three experimental groups than those in the control group(P<0.05),and the grades of traumatic proliferative vitreoretinal lesions in group A were higher than those in groups B and C(P<0.05)The grade of traumatic traumatic proliferative vitreoretinal lesions in groups B and C was not statistically significant(P>0.05);at the fourth week after surgery,α-SMA expression in the vitreous retina were higher in the experimental groups than in the control group(P<0.05);at the fourth week after surgery, the sections of the experimental group showed a higher degree of retinal proliferation than that of the control group. Conclusion:On the basis of causing penetrating eye injuries, intravitreal luminal injections of RPE,PRP,and TGF-β2 all caused traumatic proliferative vitreoretinopathy in rabbits, with vitreous injections of PRP modeling the most pronounced effect.
