Study On Expressions Of VEGF, COX-2 And CD31 In Cervical Carcinoma And Cervical Intraepithelial Neoplasia

Objective: To investigate the expressions of CD31, VEGF AND COX-2 in cervical carcinoma .cervical intraepithelial neoplasia and their significances and relationships with clinical features.Methods: Immunohistochemical technique was used to detect the signal microvessl density of CD31 and the expressions of VEGF and Cox-2 in 47cases of cervical carcinomas, 17cases cervical intraepithelial neoplasia and 15 chronic cervicitis tissues.Results: (1) The expression rates of VEGF was 55.3% ,23.5% ,6.0% in cervical carcinomas, intraepithelial neoplasia and chronic cervicitis tissues, respectively. Their relationships of expression have significant differences(p<0.01).The expression rates of VEGF was 12.5 % ,25.0 % ,40.0 % in I, II,IIIphase of cervical intraepithelial, neoplasia, and their relationships of expression has no significant differences, respectively. (p=0.798>0.05).Expression of VEGF was significantly associated with pathological grade, depth of myometrial invasion and clinical phase of cervical carcinomas (p<0.05),but was irrelevant to age of patients , pathological type and lymph node metastasis (p>0.05). (2)The expression rates of Cox-2 was 72.3%, 17.6%, 20.0%, in cervical carcinomas, cervical intraepithelial neoplasia and chronic cervicitis tissues, respectively. Its expression was significantly different along cervical carcinoma .cervical intraepithelial neoplasia, and chronic cervicitis tissues (p< 0.01).The relationships of the positive expression between cervical intraepithelial neoplasia, and chronic cervicitis tissues has no significant difference (p>0.05).The expression rates of Cox-2 was 12.5%, 25.0%, 40.0% in I , II ,IIIphase of cervical intraepithelial, neoplasia ,and their relationships of the positive expression has no significant difference, respectively (p>0.05).The positive expression of Cox-2 was significantly associated with pathological grade, phase of cervical carcinomas, (p< 0.05),but was irrelevant to age of patients ,depth of myometrial invasion , lymph node metastases, and pathological type(p>0.05). (3) The miscrovessl density was 8.72±1.28, 24.50±2.43, 48.26±3.27 respectively in chronic cervicitis tissues, cervical intraepithelial neoplasia and cervical carcinomas. relationships of their microvessl density has significant difference(p<0.05).The microvessl density was 26.27±1.62, 27.89±0.87, 28.41±1.63 respectively in I, II,III phase of cervical intraepithelial neoplasia. The relationships of those microvessl density has no significant difference(p>0.05). The microvessl density was significantly associated with phase of cervical carcinoma, depth of myometrial invasion and pathological type (p<0.05), but was irrelevant to age of patients, lymph node metastases and grade (p>0.05).(4) The relationships of the expression of VEGF and Cox-2 in microvessl density in two groups of middle in cervical carcinoma has significant difference(p<0.01).(5) The relationship the expression of VEGF and COX-2 in cervical carcinoma has significant correlation.Conclusions: (1) Expression levels of VEGF is increased with the transformation from chronic cervicitis tissues to cervical intraepithelial neoplasia to cervical carcinoma. The experiment suggests that expression of VEGF occurred in early stage of cervical carcinoma transformation, was closely related to the clinical progress of cervical carcinoma .Higher levels of it may indicate infausta prognosis for patients with cervical carcinoma. (2) Expression levels of Cox-2 is increased with the transformation from chronic cervicitis tissues to cervical intraepithelial neoplasia to cervical carcinoma, and was carefully related to clinical progress of cervical carcinoma, pathological grade and phase of cervical carcinomas. Decreased expression of Cox-2 is a useful parameter of cervical malignancy. (3) CD31 is a particular sign for the new microvessl. The signal microvessl density of CD31 was increased with the transformation from chronic cervicitis tissues to cervical intraepithelial neoplasia to cervical carcinoma, The microvessl density has no significantly differences in cervical intraepithelial neoplasia with different pathological grade, and was carefully related to phase of cervical carcinoma and depth of myometrial invasion(p<0.05). The experiment suggests that it may a useful parameter of cervical malignancy. (4) It is correlative between positive expression of VEGF and COX-2 in cervical carcinoma, and they were associated with the miscrovessl density. The experiment suggests that all of them were used to being metastasis of cervical carcinomas and form of new miscrovessl.
