ObjectiveThis study was designed to observe the expression of PTEN,VEGF and CD105in normal cervical epithelia and cervical carcinoma. Furthermore, to analyze their association with the pathological features and their relationship with the early diagnostic, development, metastasis and prognosis of the cervical carcinoma, providing a new effective index for cervical cancer diagnosis and treatment.MethodsThe expression of PTEN,VEGF and CD105was investigated by IHC in90case, including23cases of normal cervical epithelia and67cases of cervical carcinoma. Blood vessel endothelial cells were labeled by CD105and the MVD in the cervix carcinoma was assayed with weidner. The expression level of PTEN,VEGF and CD105in cervical carcinoma was analyzed against clinical pathological factors such as clinical stage,histological grade and lymphatic metastasis,moreover, study of correlation between the three in the process of development in cervical carcinoma. The expression of CD105was labeled by MVD (CD105-MVD)Results1.The percentage of VEGF,PTEN and CD105positive expression in normal cervical epithelia is13.04%、100%and1.88±2.124but in cervical carcinoma the positive percentage is76.1%、44.8%and31.49±2.820.The different of expression in cervical carcinoma and normal cervical epithelia was statistically significance(P<0.05).2.VEGF and CD105-MVD were expressed significantly higher in cervical carcinoma with lymphnode metastasis than without, the different of the expression were statistically significance(P<0.05). PTEN was expressed significantly lower in cervical carcinoma with lymphnode metastasis than without(P<0.05). The percentage of VEGF、PTEN and CD105positive expression were different in cervical carcinoma with different clinical stage, with the clinical stage increased the positive expression of VEGF and CD105-MVD was significantly higher, but the positive rate of PTEN expression decreased, the different of the expression were statistically significance (P<0.05). There were no significantly difference between PTEN、VEGF and CD105-MVD and other clinical features (different age, gross type, degree of differentiation and histological types), the different of the expression were no significance(P>0.05).3.The MVD was significantly higher in PTEN(-)、VEGF(+) cervical carcinoma than PTEN(+)、VEGF (-).Using Spearman correlation analysis, the expression of PTEN and VEGF was negative correlated in cervical carcinoma(rs=-0.270,P<0.05), the expression of CD105-MVD and VEGF was positive correlated in cervical carcinoma (rs=0.715,P<0.05), the expression of CD105-MVD and PTEN was negative correlated in cervical carcinoma (rs=-0.865,P<0.05)ConclusionThe expression of CD105-MVD and VEGF was obviously increased form normal cervical epithelia to cervical carcinoma but the expression of PTEN was obviously decreased. In the cervical carcinoma with lymphatic metastasis and higher FIGO the expression of CD105-MVD and VEGF was obviously higher than without and lower FIGO, however, the expression of PTEN was contrary. we concluded that PTEN、 VEGF and CD105may play an important role in the development and metastasis of cervical carcinoma. The CD105-MVD was increased in cervical carcinoma tissues with VEGF expression higher, which conclude that CD105-MVD can be used as a quantified index to evaluate the activity of VEGF to provide a theory foundation for the cancinoma’s biological behavior, prognosis and therapy with blood vessel. The expression rate of VEGF and CD105-MVD was increased in cervical carcinoma tissues with PTEN expression lower, PTEN protein expression decreased or absent may up-regulate the expression of VEGF and CD105expression to promote cervical cancer occurrence, development and metastasis; Elevation of PTEN and inhibition of VEGF and CD105expression may be a effective way of therapy with blood vessel for cervical carcinoma, prevent the lymphnode metastasis and improve the patient’s prognosis. Combined detection of PTEN,VEGF and CD105has important significant for cervical carcinoma’s diagnosis, therapy and assessment of prognosis.
