Objective:Exploring the auto-fluorescence spectra changes and differences of serums of patients, who have liver cancer, esophageal cancer, gliomas and lung cancer in order to provide spectral theory foundation for the research of fluorescent materials in occurrence and progress of tumors.Materials and methods:1.Subjects and sample collectingThe subjeces were cancer patients hospitalized and healthy subjects in the First Affiliated Hospital of Zhengzhou University ,Henan Province ,from October 2005 to September 2006. Collecting 3ml blood and serum from the upper centrifugal. The study collected by gender, age and biochemical indicators.2. The auto-fluorescence spectra of serum were determinedUse the RF-4500 photometer to determine groups of the auto-fluorescence spectra of serum.3. Statistical analysisUse SPSS13.0 statistical software to analyze the parameters. Independent t-test and one way ANOVA were used to analyze the peak position, the height,the area and the ratio between the height of the auto-fluorescence spectra in serum among the normal control with different cancers. One way ANOVA was used to analyze the peak position, the height ,the area and the ratio between the height of the auto-fluorescence spectra in serum among different liver disease and lung cancer patients with different chemotherapy cycle.Results:1. Stimulated by 300nm ultraviolet radiation, the auto-fluorescence spectra of serum have six strong peak in 200-800nm fluorescence wavelength range , reflect more informative of the difference in tumor patients and normals. The auto-fluorescence spectra of serum unchanged basically within six hours.2. Hepatitis B group have the potential to move to the violet band at 448nm, the fluorescence height and area of hepatitis B are both obviously heightened at the place of 603nm and those of the hepatocirrhosis patients were higher at 302nm , liver cancer group have the potential to move to the red band at the place of 648nm and the ratios between the height were higher at 603nm and 648nm, and the differences were remarkable compared with the normal group(p<0.05).3. The fluorescence height and the area of esophageal cancer were higher at the point of 302nm, 336nm, 448nm and 648nm,the ratios between the height were higher at 302nm and 336nm. The differences were remarkable compared with the normal group(p<0.05).4. Those of the Gliomas have moved to the red band at the place of 448nm , the fluorescence height were higher at the place of 302nm, 336nm, 448nm, 514nm, 603nm and 605nm , the ratios were higher at 302nm and 603nm , the fluorescence areas were broader at the place of 302nm, and the differences were remarkable compared with the normal group(p<0.05).5. Those of lung cancer patients before chemotherapy have the potential to move to the violet band at 336nm and 509nm, the fluorescence height at the place 302nm, 448nm, 514nm, 603nm and 648nm were higher and the fluorescence area at the place of 302nm, 448nm and 603 nm were higher; Lung cancer after first cycle of chemotherapy have potential to move to the violet band and the fluorescence intensity were lowered at the place 336nm ; Lung cancer after second cycle of chemotherapy have potential to move to the violet band at the place of 336nm, the fluorescence height were higher at the place of 448nm and the fluorescence area were higher at the place of 336nm and 448nm;The differences were remarkable compared with the normal group(p<0.05).Conclusion:1. The auto-fluorescence spectra at the place of 648nm may be the characteristic auto-fluorescence spectra of liver cancer, the research of fluorescent materials's structure at 648nm can provide clues for liver cancer's incidence studies .2. The peak height and area at the place of 302nm,336nm,448nm and 648nm may be auto-fluorescence spectra's characteristic characterization of esophageal cancer, according to the fluorescence intensity is proportional to the quantity of fluorescent materials, this surmised that the incidence of esophageal cancer may be related to the increase of fluorescent material at the place of 302nm, 336nm,448nm and 648nm, the further study of these substances quantitative can provide clues of the mechanism for the incidence of the esophageal cancer.3. The characteristic features of the glioma patients's auto-fluorescence spectra:the red shift of peak position at the place of 448nm, fluorescent, the i height increased at the place of 302nm,336nm,448nm,514nm,603nm and 648nm, the ratios were higher at 302nm and 603nm , these features was in-depth research can provide spectra's arguments for the incidence of glioma and early screening .4.The auto-fluorescence spectra's changes of lung cancer after chemotherapy reflect the effect of chemotherapy drugs on tumor, with the depth of research, the determination of auto-fluorescence spectra's changes before and after chemotherapy is expected to evaluate the effect of chemotherapy for spectroscopic arguments.
