Objective To study the antiemetic efficacy of the extraction of ginger and to investigate potential mode of action, in this case, we'll have the chance to develop our traditional herbs into neoteric naturally occurring drugs.Metheds 1. To prepare the ethanol extraction of ginger and ginger juice.2. To establish the emetic model in rats. The rats were randomly divided into various groups (six animals per group), the animals were administrated different dose of cisplafin , apomorphine, rotated(100 r-min-1, 1h)and copper sulfate, then the consumption of kaolin was measured. According the consumption of kaolin we can know whether the model is succeed. Then the animals were given antiemetic reagent and the antiemetic effect were evaluated.3. on the basis that the emetic model was successful got, the minks were randomly divided into various groups (six animals per group), the animals were given different dosage of the ethanol extraction of ginger, ginger juice and the positive drug ondansetron, on the following 6h and 24h, the antiemetic were observed.4. Via immunohistochemistry test we can measure the contents of SP on the tissue of sinus ventriculi of minks and rats from the control groups and other groups, so we can observe the relationship between the expression of SP and the test.5. For the PDD(2.5mg·kg-1 )groups, after 24h the administration of PDD and antiemetic reagent, the rats were luxated cervical vertebrae and the adbo were cut and a piece of ileum about l cm were got for testing the contents of 5-HT and 5-HIAA by HPLC-ECD.Results 1.With cisplatin (2.Smg·kg-1), apomorphine (3.2mg·kg-1 )and rotation situation(100r·min-1 1h), the rats expressed eating non—nutrition substances kaolin, so- called pica, the contents of exps significantly different from the control group. P<0.012.the ethanol extraction of ginger and ginger juice can inhibited the pica behavior in rats induced by vomito such as cisplatin, apomorphine and rotation.3. The ethanol extraction of ginger and ginger juice can against emetic induced by vomito such as cisplatin, copper, sulfate, apomorphine and motion sickness In minks, the emetic latency was increasedthe No. of retch episodes and the No. of emetic episodes were decreased compared with the control group (P<0.05, P<0.01 )4. The result of mmunohistochemistry test revealed that, compared with the positive drug ondansetron group, the count of the postive neurofibra in the muscular layer of sinus ventriculi in antiemetic reagent groups was significantly diminished and the tinction was light.5. The result of HPLC manifests that the release of 5-HT induced by PDD can be resisted by the antiemetic reagent ginger contrast to the control group. The cont of 5-HT and 5-HIAA show a negative correlation.Conclusion 1. The rats' pica model are successful established and estimated. The pica behavior can be opposed by The ethanol extraction of ginger and ginger juice.2. The ethanol extraction of ginger and ginger juice can significantly inhibit the emetic in minks.3. The potential mode of antiemetic action correlated with SP and 5-HT.
