Objective:To study the expressive correlation between OPN and VEGF in breast cancer. To explore expressive effect of OPN, VEGF and their unit in the metabasis and recurrence of breast cancer. To investigate the relationship between OPN and VEGF, their correlation with ER, PR, C-erbB-2, P53 and the clinical and pathological factors in breast cancer.Materials and methods:Took 70 cases from Xiangya Hospital Department of Breast from January 1996 to October 2001 randomly, made out their survival time by investigations, including 40 cases with recurrence or metabasis in 5 years(Group A), 30 cases without recurrence or metabasis in 5 years (Group B) and 10 cases with breast fibroid tumor (Group C ) and 15 cases normal breast tissue(Group D).Their clinical stage wasⅠstage(5),Ⅱstage(56) andⅢstage(9) according to AJCC 2003 clinical stage standand. All were female, and their age were from 26~74, the median age was 44.5. A11 cases were operated and tested by pathologic diagnosis. All material perfected, the observers were 100%. The initial observed time beginning operation day, the last observed time was on October 2006. The median observed month was 50.50.The expression of OPN and VEGF was studied in 70 patients with primary breast cancer, 10 patients with fibroid tumor and 15 patients with normal breast tissue by immunohistochemical staining technique. All statistics analysis was used by SPSS 11.5 for window. Single factor was used byχ~2 test of crosstable, spearman's correlation and Log-rank test of survival time to analyze prognostic factors in primary breast cancer. Kaplan-Meier method was used to analyze the relationship of their expression with prognosis. The relationship of two protein expression, the clinical and pathological parameters and prognosis were analyzed.Result: (1)The expression positive rate of OPN was 72.86%(51/70) in 70 patients with primary breast cancer, and only 2 positive expression of 10 breast fibroid tumor and 4 positive expression of 15 normal breast tissues. There was significantly different between breast cancer, breast fibroid tumor and normal breast tissue(P<0.001).(2)The expression positive rate of VEGF was 70.00%(49/70) in 70 patients with primary breast cancer, and only 3 positive expression of 10 breast fibroid tumor and 4 positive expression of 15 normal breast tissue. There was significantly different between breast cancer and breast fibroid tumor and normal breast tissue(P<0.001).(3)The date of our study, inχ~2 test, showed that the expressions of OPN, VEGF and their union were statistically significant different between group A and group B(P=0.029, 0.009, 0.045). The results of survival analysis with Kaplan-Meier method showed negative OPN, VEGF and their union expression exhibited a significantly better prognosis than their positive expression(P=0.0286, 0.0010, 0.0099).(4)OPN positive expression was significantly positive correlated with that of VEGF(P<0.05, r=0.421); and not correlated with that of ER, PR, C-erbB-2, P53, but not the clinical and pathological factors such as ages, pathological, neoadjuvant chemotherapy and so on (all of P>0.05). However OPN positive expression was correlated with P53 inχ~2 test, but not in spearman's correlation.(5)VEGF positive expression was significantly correlated with that of P53 and C-erbB-2(P<0.05, r=0.327, 0.362); but not correlated with that of ER, PR and others factors(all of P>0.05).Conclusions:(1)The expression positive rates of OPN and VEGF between breast cancer, breast fibroid tumor and normal breast tissue were significantly different(P<0.05). OPN and VEGF may be correlatied with the development of breast carcinoma.(2) OPN and VEGF were associated with the prognosis of breast cancer. The patients with the union expression of OPN and VEGF had worse prognosis, which may be provide some new and valuable targets as molecule targeted therapy for breast carcinoma. (3) OPN was positive correlated with VEGF but not with ER, PR and others factors. However it's relation with P53 was not sure. This needs more hakeems to study deeply.(4) VEGF was positive correlated with many genes associated with the prognosis of the breast cancer, including P53 and C-erbB-2. But it was not associated with ER, PR and the clinical and pathological factors.In breast carcinoma the overexpression of OPN and VEGF could be an independent prognosticator. Detecting their expression may be helpful in selecting the high risk cases for further treatment.
