Black-bone silky fowl (Gallus gellus demesticus brissen) is a kind of bantambreed which native to South China, Taihe, Jiangxi Province. The original black-bonesilky fowl (BSF) is well known in the orient and has been credited with famousmedicinal and health-promoting values for more than a thousand of years. As one ofthe important bioactive components of BSF, phospholipids (PL) has been studiedprofoundly in this paper, it will provide theoretic foundation for the integratedexploitation of BSF and its bioactive components. The main studies and results in thepaper are mentioned as follows:The Molybdenum blue colorimetry for determination of total phosphorus in PLfrom BSF was described. The contents of PL from muscles of BSF were 5.11 mg/g infresh meat, 22.0 mg/g in dry meat and 5.23%of total lipids.Mixed solvent-ultrasonic extraction was the most effective extraction method bycomparison with other different methods. Orthogonal experiment was carried out andthe optimum technical parameters were obtained: 3:1 of the ratio of hexane toisopropanol, 1:7 of the ratio of material to solvent, 30℃of reaction temperature for40 min, extracts two times. The verifying experiment showed that the comparativeextraction ratio of PL reached 85.29%.A method combined silica gel solid phase extraction with TLC was establishedto determine the PL classes in BSF. The extraction conditions were found: 0.8 ml of400 mg/ml sample, diluted with 4 ml chloroform, followed by 8 ml methanol. Therecovery rate of PL can achieve 95%. The developing solvent of TLC was composedof chloroform/methanol/acetic acid (6.07:2.43:1.5, V/V) and 8cm was the properdeveloping distance. It found that the four distinctly separated spots were LPC, PC,PI and PE according to the standards.A simple and convenient HPLC method was developed to determine the PL fromBSF. The separation was conducted on a MN nucleosil 100-5 column withacetonitrile/methanol/water (8:2:1, V/V) as mobile phase. Satisfactory linearity and recovery value demonstrated the method was fit for the PL analysis from BSF.The fatty acids of PL were identified and analyzed by GC-MS. 8 fatty acids werefound in total PL which characterized by high contents of polyunsaturated fatty acidsand arachidonic acid. All the 8 fatty acids were detected in PC and PE, Eicosatrienoicacid was not detected in LPC, palmitic acid and Eicosatrienoic acid were not detectedin PI. PC,PE,LPC had high contents of Stearic acid and arachidonic acid, the contentof palmitoleic acid in accounted for 41.95%of total fatty acids of PI.Effect of PL from BSF on ATPase activity of Yin-deficiency rat was studied inthe paper. The results showed that PL from BSF had Yin nourishing effect for it cansignificantly lower the activity of Na+, K+-ATPase and Ca2+, Mg2+-ATPase ofYin-deficiency rat.
