Objective:To investigate the expression of NF-kB,MMP-9 and TGF-β1 in the placental of patients with hypertensive disorder complicating pregnancy,their cooperative role in hypertensive disorder complicating pregnancy.Methods:We collected all subjects from Sep 2005 to Mar 2007 in the department of Obstetrics and Gynecology,First Clinical Medical college,Shanxi Medical University.The clinical subjects were divided into three groups:①gestational hypertension:16 patients;②pre-edampsia:44 patients;③the normal pregnant women:20 patients.Immunohistochemical methods was applied to observe the expression of NF-kB,TGF-β1 and MMP-9 in the placental of patients with hypertensive disorder complicating pregnancy.Results:(1)The NF-kB in the placental was activated,At the same time,the expression degree of NF-kB was correlative with degree of hypertensive disorder complicating pregnancy.(2)TGF-β1 was found in syncytiotrophoblast and cytotrophoblast and the mean density of TGF-β1 in hypertensive disorder complicating pregnancy group was significantly higher than that of normal group in hypertensive disorder complicating pregnancy.(3)MMP-9 were expressed in trophoblast or vessel wall of villous stroma or decidual tissue.In moderate and severe pathogenesis of hypertensive disorder complicating pregnancy,the expression degree of MMP-9 was correlative with degree of hypertensive disorder complicating pregnancy.(4)The results were different among four groups was significant(P<0.05);The expression of NF-kB and MMP-9 took on negative correlation significantly(r=-0.272,P<0.05);The expression of NF-kB and TGF-β1 took on positive correlation significantly(r=0.275,P<0.05);The expression of TGF-β1 and MMP-9 took on negative correlation significantly(r=-0.49,P<0.01)Conclusion:It suggested that the activation of NF-kB,TGF-β1 and MMP-9 in the placental with hypertensive disorder complicating pregnancy and abnormal expressions of NF-kB,TGF-β1 and MMP-9 in placenta at maternal-fetal interface are related to the pathogenesis of hypertensive disorder complicating pregnancy and typical pathological change in placent.
