Objective:Hypertensive disorder complicating pregnancy(HDCP) is themost common obstetric complications, including gestational hypertension,preeclampsia, eclampsia, chronic hypertension complicated by preeclampsia,and pregnancy with chronic hypertension, gestational hypertension,preeclampsia, eclampsia is unique to pregnancy disease.Gestationalhypertension disease is a systemic disease, in order to appear after20weeksof gestation hypertension, proteinuria, edema, etc as the main characteristics,and may be accompanied by multiple organ function damage or functionfailure, serious when can appear hyperspasmia, coma, fetal growth restriction,cerebral hemorrhage, and even death, and the incidence of the disease in ourcountry is9.4%~10.4%, foreign coverage is7%~12%, seriously threatensthe maternal and child health, is the cause of maternal death and one of theleading causes of perinatal deaths.In recent years, many scholars at home andabroad to a lot of work on HDCP, however, its exact etiology andpathogenesis has not been fully elucidated.Now agree that the onset of HDCPis originated in the placental pathological physiology change, thesepathological physiological change is the result of joint action by manyfactors.At home and abroad in recent years, a growing number of studiessupport the sertoli cell apoptosis increased, infiltration capacity is impaired,invades the uterine spiral artery too shallow, placental vascular physiologicalrecasting caused by obstacles such as shallow placenta implantation is the keyfactor of pregnancy hypertension disease.Nourish cells cannot invade theuterus place, infiltrating sertoli cell infiltration shallow, shortage and uterinespiral artery recasting failure can often in the placenta tissue of patients withgestational hypertension disease is observed.IMP3(insulin-like growth factorⅡ mRNA binding protein3) is one of the members of the insulin-like growth factor Ⅱ mRNA binding protein family, is a kind of secretory proteins thatcan combined with insulin-like growth factors (IGFs), can unify highlyspecific encoding the insulin-like growth factor Ⅱ mRNA, participate in thelocation of the insulin-like growth factor Ⅱ mRNA, by adjusting theinsulin-like growth factor Ⅱ mRNA stability of transportation, translation,and to participate in the regulation of cell polarity, movement, andproliferation activity, is closely related to the invasion and metastasis ofcells.According to the relationship of IMP3with invasion and metastasis ofcells, presumably, its abnormal expression may changes of sertoli cellinfiltration, plays an important role in the happening and the development ofhypertensive disorder complicating pregnancy. This study using western blotmethod respectively testing IMP3expression in placenta tissues of patientswith pregnancy hypertension disease and normal pregnant women, to explorethe placenta tissue of patients with pregnancy hypertension disease expressionand its clinical significance, in order to find cause of gestational hypertensiondisease for clinical provides a new basis, and improve the early diagnosis ofpregnancy hypertension disease and early treatment.Materials and Methods:Specimens selected from October2010to October2011in the maternityof The Second Hospital of He Bei Medical University.The normal pregnantwomen and the patients with hypertensive disorder complicating pregnancywere120cases. Select28-34weeks of pregnancy in patients with HDCP30cases as the fist experimental group, normal pregnant women for the first30patients in the control group; Gestational age more than equal to34weeks ofpregnancy hypertension patients for the second group30cases, a normalpregnancy for the second group of30cases of women.Gestationalhypertension disease diagnosis according to the people’s medical publishinghouse, Jie Le editor of journal of 《obstetrics and gynecology》(7th edition)standard, the pregnant women were selected as the first child, the averagegestational age, age, there was no statistically difference, without the historyof essential hypertension, diabetes, heart disease and kidney disease, thyroid dysfunction, etc.Samples were drawn from maternal placenta placenta villisurface near the umbilical cord root tissue, and keep away from thehemorrhage, necrosis and calcification area.After the extraction of totalprotein in the placenta tissue,the method of western blot was use to measurethe IMP3expression level in placental tissues of the two groups.Analysis ofexperimental results with UVP software, the experimental data to IMP3stripIOD values and GAPDH expressed by the ratio of the IOD values in the band.All of the experimental data used SPSS13.0statistical software for dataprocessing, measurement data to x±s said, differences between the twogroups in two independent samples nonparametric tests.The inspectionstandard was a=0.01, P<0.01as a statistically significant difference.Results:1Gestational age28to34weeks of gestation hypertension diseaseIMP3protein expression in placenta tissue is obviously lower than normalpregnant women, and the difference was statistically significant(P<0.01).2Gestational age less than34weeks of gestation hypertension diseaseIMP3protein expression in placenta tissue is obviously lower than normalpregnant women, and the difference was statistically significant(P<0.01).3Gestational age28to34weeks and gestational age less than34weeksof gestation hypertension disease IMP3protein expression in placenta tissuescompared quantity, there was no statistically significant difference(P>0.05).Conclusion:IMP3in the placental tissue of patients with hypertensivedisorder complicating pregnancy significantly lower than normal expressionpregnant women, prompting state of sertoli cell, placenta implantation isassociated with hypertension during pregnancy the happening of thedisease,suggest that the expression of IMP3in the placental tissue reduce maybe associated with hypertension during pregnancy the happening of thedisease. The early onset and late onset in patients with hypertensive disordercomplicating pregnancy placenta tissue IMP3expression had no significant difference.
