Object: Investigate the meaning and value of gene of phosphate and tension homology deleted onchromosome ten, PTEN mutation in endometrial carcinoma, the endometrial hyperplasia and the normalendometrium.Methods: Forty-three endometrial carcinoma samples, twenty-seven endometrial atypical hyperplasiasamples, twenty-four endometrial complex hyperplasia samples, twenty-seven endometrial simplexhyperplasia samples, and eleven normal endometrium samples were collected, the mutations of exon 5 andexon 8 of PTEN gene were examined by polymerase chain reaction-single strand conformationpolymorphism (PCR-SSCP), the results associated with clinical pathological features were analyzed.Results: In encometrial carcinoma, the rates of muataion of PTEN were 33.3%, which were significantlyhigher than thata of endometrial hyperplasia and normal endometrium. The rates of mutation of PTEN inendometrial atypical hyperplasia were 11.1%, endometrial complex hyperplasia were 8.3%, endometrialcomplex hyperplasia were 7.4%, normal endometrium were 0%.(1) There were significant difference of therates of PTEN mutation in encometrial carcinoma, endometrial hyperplasia and normal endometrium bystatistics(X~2=14.475, P=0.006).(2) There were no significant difference of the rates of PTEN mutationbetween endometrial hyperplasia and normal endometrium by statistics, and endometrial hyperplasiathemselves share the same condition.(3) There were no significant difference of the rates of PTENmutation between endometrial adenocarcinoma and other types of histological types (X~2=0.875, P=0.383).(4) There were no significant difference of the rates of PTEN mutation between the surgery-pathologicalstageⅠand stageⅡ~Ⅲby statistics (X~2=0.077, P=1.000). (5) There were no significant difference ofthe rates of PTEN mutation in histological grade by statistics (X~2=0.244, P=0.885). (6) There were nosignificant difference of the rates of PTEN mutation between the superficial myometrium involvement anddeep myometrium involvement by statistics (X~2=0.525, P=0.719).Conlusions: (1) Both of endometrial carcinoma and endometrial hyperplasia might develop PTENmutation, which is higher in endometriat carcinoma, however, there is an increasing trend of PTENmutation with the severity of endometrial hyperplasia. (2) There were no relationship between PTENmutation and the histological types, surgery-pathological stage, histological grade and myometriuminvolvement, however, there is a decreasing trend of PTEN mutation with the increased histological gradeand surgery-pathological stage. (3)It need to add more samples in the research on PTEN mutation point indifferent cases, different endometrium to know the risk factors between endometrial carcinoma and PTENmutation. (4)The examination of PTEN mutation is benefit for the early diagnosis and can provide thereference for the diagnosis of pathologist and the management of clinical gynecologist.
