Objective: To study the pattems of the expression of S-phase kinase associated protein2 (SKp2) and Motility-related protein -1 (MRP-1/CD9) in human laryngeal cancerand different regions adjacent tumor to investigate the relation between different clinicalstaging, pathological grade and expression of SKp2 and MRP-1/CD9 and to investigate thesafe surgical margin.Methods: The paraffin-embeded specimens from 38 laryngeal squmous cell carcinoma(LSCC) were chosen from all who underwent primary surgical resection at our departmentbetween 2001 to 2002. Each patient's specimens was ranged and studied in different regionsof larynx including carcinoma, adjacent mucosa which was 2mm, 5mm, 10mm away fromthe tumor. Ten mucosa of polyp of vocal cord was the controal groups. Monoclonalantibodies specific for SKp2 and MRP-1/CD9 were used for immunohistochemical detectionof the patient's expression in those tissue section. The expression level of SKp2 andMRP-1/CD9 protein were examined and the result were studied with statisticalsoftware(SPSS) in order to investigate the relation between different clinical staging,pathological grade and to compare the different expression of them in human laryngealcancer and different regions adjacent tumor.Results: The positive expression rate of SKp2 was 50.00% in LSCC, 2.63% incontronal groups and the statistical difference was significant. The rate respectively was44.74%, 23.68 %, 7.89% in different regions of larynx adjacent mucosa which was 2mm,5mm, 10mm away from the tumor. The positive expression rate was 2.63% in contronalgroups. The results also showed that SKp2 proteins decreased as the tissue progressed insequence from carcinoma tissues to 2mm, 5mm, 10mm distant mucosa adjacent carcinomaand groups. The statistical differrence between carcinoma tissues and edge group (5mm,10mm) was significant(P<0.05) respectively, but it was not significant between carcinomatissues and tissues near the tumor(P>0.05) and it was not significant between 2mm tissuesand 5mm tissues. The positive product of SKp2 was mainly localized on cellular nucleus.The positive expression rate of MRP-1/CD9 was 49.20% in LSCC, 100.00% in contronalgroups and the statistical difference was significant. The rate respectively was 61.05%,89.47%, 100.00% in different regions of larynx adjacent mucosa which was 2mm, 5mm,10mm away from the tumor. The positive expression rate was 100.00% in contronal groups.The results also showed that MRP-1/CD9 proteins increased as tissue progressed in sequencefrom carcinoma tissues to 2mm, 5mm, 10mm distant mucosa adjacent carcinoma andcontronal groups. The statistical difference between carcinoma tissues and edge group(5mm,10mm) was significant(P<0.05) respectively, but it was not significantbetween carcinoma tissues and tissues near the tumor(P>0.05) and it was not significantbetween 2mm tissues and 5mm tissues. The positive product of MRP-1/CD9 was mainlylocalized on cell membrane.Conclusion: SKp2 and MRP-1/CD9 may act as a reference index in judging thebiological speciality of LSCC. It is appropriate to regard 5mm or 5mm~10mm away fromtumors as resection margins for surgical treatment of laryngeal carcinoma.
