Objective Laryngeal carcinoma is high frequency one in all head and neck malignant tumor. It is important to study the relation between expression of PTEN in laryngeal carcinoma and its' clinical characteristics for knowing biological action of laryngeal tumor.It is quite important to choose a suitable wide resection margin because the remains of cancer cells of resection margin means failure of treatment. Nowdays, morphology is still a primary standard of estimating surgical margin. With development of molecular biology and more studying of neoplasm occurrence and progress, it is valuable to estimate safe resection margin with molecular biology method. To study the patterns of the expression of PTEN in human Laryngeal cancer and different regions adjacent tumor to investigate the relation between different clinical Staging , pathological grade and expression of PTEN and to study the range of safe surgical margin. It can instruct clinical train in certainly degree.Methods The paraffin-embeded specimens of 38 laryngeal squmous cell carcinoma(LSCC) patients were randomly chosen from all whose underwent primary surgical resection at our department between 2001 and 2002..Each patient's specimens were ranged and studied in different regions of larynx including carcinoma, adjacent mucosa which was 2mm,5mm,10mm away from cancer. Ten mucosa of polyp of vocal cord was comparative groups. Monoclonal antibodies specific for PTEN were used for immunohistochemical detection of each protein in those tissue section. The expression level of PTEN protein were examined and the result were were studied using statistical software in order to investigate the relation between different clinical Staging , T, pathological grade and expression of PTEN and to compare different expression of PTEN in in human Laryngeal cancer and different regions adjacent tumor.Result The positive expression rate of PTEN was 60.53% in laryngeal carcinoma tissues, 100.00% in comparative groups and the statistical difference was significant. The rate respectively was 71.05. 89.47%. 100. 00% in different regions of larynx adjacent mucosa which was 2mm,5mm,10mm away from cancer. The positive expression rate was 100% in comparative groups. The results also showed that PTEN proteins increased as tissue progressed in sequence from carcinoma tissues to 2mm,5mm,10mm distant mucosa adjacent carcinoma and comparative groups. The statistical differdnce between carcinoma tissues and edge group(5mm, 10mm) was significant(P1<0.05, P2<0.05) respectively, but it was not significant between carcinoma tissues and tissues near the tumor(P>0.05). The stained cells for PTEN were normal cells. The positive product of PTEN was mainly localized in cellular nucleus.Conclusion PTEN was tumor suppressor gene. PTEN may act as a reference index in judging the biological speciality of LSCC. Lower expression of PTEN was may play an important role in LSCC. It is appropriate to regard 5mm away from tumors as resection margins for surgical treatment of laryngeal carcinoma.
