Objective: To investigate the antineoplastic role and its mechanism ofGlycosaminoglycan from scallop shirt(SS-GAG) in vitro and in vivo inhibitory tumor experiment.Methods:1. In vitro experiment: In vitro, four tumor cell lines (LOVO,HeLa,SMMC 7721,A549)were cultured with SS-GAG at the final concentrations(500,200,100,50,25μg·ml-1).Following incubation, MTT chromatometry was used to measure the effect of SS-GAG on theproliferatory activity of tumor cells. 2. In vivo experiment: 1)Mouse portability sarcoma 180 ascitescarcinoma model was set up to observe the survival time in 30 days and calculate life prolongation rate.2) Mouse portability sarcoma 180 solid tumor model was established to observe the antineoplasticactivity of SS-GAG and Its effect of the auxodrome, the index of spleen and thymus, the liver weight,the leukocyte count. 3. Mechanism study: 1) MTT method was used to observe the effect of SS-GAGon the phagocytic activity, killing and wounding activity of peritoneal macrophage and observe theeffect of SS-GAG on the proliferate-ion of splenic lymphocyte and NK cell killing activity of S 180oncocyte immunized mice, 2) A lot of biochemistry methods including xanthine oxidase method andTBARs were used to study the effect of SS-GAG on the oxidation production of lipoproteinmalondialdehyde (MDA) and activity of superoxide dismutase (SOD) and total antioxygen activity inserum of S 180 oncocyte immunized mice.Results: 1. In vitro experiment: The glycosaminoglycan from scallop shirt(SS-GAG)couldsignificantly inhibit the growth of lovo,HeLa,A549 cell lines in a dose-dependent manner, and therewas weak effect on the growth of SMMC7721 cell line 2. in vivo experiment: The SS-GAGsignificantly lengthened the survival time of the mice transplanted in the abdominal cavity with S 180humor, inhibited the growth of S 180 humor in mice. Enhance the antineoplastic activity and lightenthe toxicity of CTX. 3. Mechanism study: 1)SS-GAG could significantly enhance celiacmacrophagocytic and cytotoxic activity of S180 oncocyte immunized mice, promote proliferation of spleno cytes induced by suboptimal concentration of mitogen concanavalin A, and reinforce NK cellkilling activity of mice treated by SS-GAG. 2)The SS-GAG significantly enhanced the serum's totalantioxygen activity, promoted the activity of SOD, decreased the malondialdehyde(MAD) content.Conclusion: The glycosaminoglycan from scallop shirt(SS-GAG) has a depressant effect onexperiment tumor model; the mechanism of anti-humor effects of the glycosaminoglycan from scallopshirt(SS-GAG) can attributed to improve function of the immune organs and enhance anti-oxidativecapacity in humor-bearing mice.
