The expression and significance of human pituitary tumor transforming gene protein in the endometrial carcinomaObjectiveTo study the expression and significance of pituitary tumor-transforming gene(hPTTG)protein and its relationship with the expression of epithelia-cadherin(E-cad) in endometrioid adenocarcinoma by to exam the expression of hPTTG protein in normal endomerium, simple hyperplasia of endometrium, atypical hyperplasia of endometrium and endometrioid adenocarcinoma.MethodsImmunohistochemistry of PV-9000 polymer detection systerm was used to detected the expression of hPTTG and E-cad protein in 46 cases of endometrioid adenocarcinoma with different clinical pathologic characteristics. 8 cases of atypical proliferation of endometrium, 11 cases of simple proliferation of endometrium, 17 cases of normal endometrium including 8 cases of secret stage and 9 cases of proliferative stage, then compared and analyzed these values among each other.Results1. The expression of hPTTG protein in endometrioid adenocarcinoma is overpressed(67.4%), and hPTTG protein in atypical hyperplasia of endometrium(5/8) was significantly higher than in simple hyperplasia of endometrium(18.2%)and normal endometrium tissues(17.6%), there was a significant difference(P<0.01). The expression of hPTTG protein in endometrioid adenocarcinoma was related to surgical-pathological stage, the grade of cellular differentiation, myometrial infiltration depth and lymphatic metastasis(P<0.05, respectively).2. Normal expression of E-cad protein was found in all of 17 cases of normal endometrium tissue. The abnormal expression rate of E-cad protein in atypical hyperplasia of endometrium and endometrioid adenocarcinoma were higher than in normal endomerium and simple hyperplasia of endometrium(P<0.05). The abnormal expression rate of E-cad protein in endometrioid adenocarcinoma was related to surgical-pathological stage, the grade of cellular differentiation grade, myometrial infiltration depth and lymphatic metastasis.3. The abnormal expression rate of E-cad protein in endometrioid adenocarcinoma tissues with positive expression of hPTTG protein(71.0%)were higher than in those with negative expression of hPTTG protein(20.0%), the difference is significant(P<0.01). The abnormal expression of E-cad protein and the expression of hPTTG protein in endometrioid adenocarcinoma present positive correlation(r=0.486).Conclusions1. The expression of hPTTG protein in endometrioid adenocarcinoma was overpressed. The positive expression rate of hPTTG protein in endometrioid adenocarcinoma was related to surgical-pathological stage, the grade of cellular differentiation grade, myometrial infiltration depth and lymphatic metastasis, hPTTG protein promoted the carcinogenesis and development of endometrioid adenocarcinoma.2. The expression of E-cad protein in endometrioid adenocarcinoma was decreased, The abnormal expression rate of E-cad protein in endometrioid adenocarcinoma was related to surgical-pathological stage, the grade of cellular differentiation grade, myometrial infiltration depth and lymphatic metastasis. The abnormal expression of E-cad protein promoted the infiltration and metastasis of endometrioid adenocarcinoma.3. The abnormal expression of E-cad protein and the expression of hPTTG protein in endometrioid adenocarcinoma presented positive correlation.
