Objective It has been suggested that pituitary tumor-transforming gene(PTTG),a novel oncogene,has played an important role in many human tumor.However ,its precise role has not been known in uterine leiomyomas and the development of the tumor.In this study ,the expressions of PTTG protein were detected and the relationship with the clinical features,the expression of basic fibroblast growth factor (bFGF) protein and c-myc protein were analyzed in uterine leiomyomas.Methods Expressions of PTTG,bFGF and c-myc protein were examined by immunohistochemistry SP method in 60 cases of uterine leiomyomas(32 cases of solitary hysteromyoma,28 cases of multiple hysteromyoma),and compared with clinical characteristics ,such as the size and the situation of the tumor,the age of the patient and expression of protein bFGF and c-myc. 12 cases of normal myometrium were investigated as control group.Results 1. PTTG was expressed in hysteromyoma tissues and normal myometrium tissues,the positive rate were 84.1% and 16.7%.The expression of PTTG protein in uterine leiomyomas was significantly higher than in normal myometrium tissues (P<0.001).2. The positive rate of PTTG was strongly associated with the volume of the uterine leiomyomas.The expression rate of PTTG protein was stronger in the big hysteromyoma(the diametor >5cm) 92.3% than in the small hysteromyoma(the diametor≦5cm) 72.2% (P=0.011) .There was no relationship between PTTG protein expression and age,situation of the tumor or type(P>0.05).3. The expression rate of PTTG protein in uterine leiomyomas tissues with the bFGF protein was significantly positive correlate (r=0.718, P<0.01 ) .And the expression rate of PTTG protein in uterine leiomyomas tissues with the c-myc protein was also significantly positive correlate (r= 0.718, P<0.01) .Conclusions 1. The expression of PTTG protein in uterine leiomyomas was significantly higher than those in normal myometrium tissues.The results suggest that the overexpression of PTTG may play an important role in the genesis of hysteromyoma.2. Over expression of PTTG protein were related to clinical characteristics,such as the volume of the uterine leiomyomas .The results suggest that the overexpression of PTTG may play an important role in the development of hysteromyoma..3. There was a relationship between PTTG expression and the expression of bFGF or c-myc protein.PTTG together with bFGF and c-myc may play an important role in the genesis and the development of hysteromyoma.
