Objective: To explore the relationship of adiponectin gene polymorphism with atherosclerotic cerebral infarction (ACI) in Han population in Hunan Province, and the possible influence of these SNPs on blood fat and blood pressure.Methods: Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) and DNA sequencing were performed to identify the distributions of allele and genotype frequencies of SNP45T/G polymorphism and SNP276G/T polymorphism in adiponectin gene in 155 patients with ACI and 149 healthy controls. Serum total cholesterol (TG) and triglyceride concentrations (TC) were determined by an enzymatic method. High-density lipoprotein cholesterol (HDL-C) was also measured by an enzymatic method after heparin and calcium precipitation. Low-density lipoprotein cholesterol (LDL-C) was estimated by using formula (LDL-C)=(TC)-(HDL-C)-(TG/5).Results:1. Single-point genotypes and alleles distributions in SNP45 of the adiponectin gene were not statistically different between the ACI and control groups.2. The distributions of genotypes and alleles of SNP276 both displayed significant difference between ACI and control groups, along with the T allele being significantly increased in the ACI group (0.341) compared with the control group (0.262). Moreover, in haplotype estimation analysis, there was a higher frequency of the haplotype TT in the ACI group (13.5%) than in the control group (5.3%), the carriers of the TT genotype had a higher risk of developing ACI.3. Carriers of the 276TT homozygote exhibited significant higher serum TG as compared with 276GG homozygote carriers in ACI subjects. Mean TG concentration among the 276TT subjects was 2.16 mmol/1 and that in subjects with the 276GG genotypes was 1.54 mmol/l.Conclusions:1. The 276G/T polymorphism of APN gene may correlate with ACI in Han population in Hunan province. There is no relationship between the SNP45T/G polymorphism of APN and ACI.2. The 276G/T polymorphism of APN gene may be associated with blood fat level. Carriers of the 276TT homozygote may be involved in high level of TG. The roles of adiponectin gene polymorphism in ACI need to be studied further.
