Objective: Pancreatic carcinoma is one of the commonmalignancies of digestive system. The reaction to the routine therapysuch as operation, chemotherapy ,radiotherapy is not so sensitive. The 5years survival is so poor that it is very necessary to find new therapymethods. Anti-sense gene-therapy as a new strategy is hopeful.Methods: Research expression of c-myc proteinum and thechanges of cell cycle and apoptosis rate in pancreatic carcinoma cells(PANC-1) after c-myc ASODN transfection, by using suchexperiment technique as cell culture, immunohist ochemistry,flow cytometry. Detection the effect of c-myc ASODNtransfection combine with 5-FU rate of growth inhibiting of Pancreaticcarcinoma cells (PANC-1) to 5-FU by the colorimetric MTT.Results: Detection expression of c-myc proteinum may decrease inpancreatic carcinoma cells (PANC-1) after c-myc ASODN transfection.and can inhibit cells from G1 phase into S phase, induce cells apoptosisAnd the effect of c-myc ASODN combine with 5-FU can increase theapoptosis sensitivity of Pancreatic carcinoma cells (PANC-1) to 5-FU by the colorimetric MTT.Conclusions: It can be conclude that c-myc ASODN is hopefulto be a efficient therapy method of pancreatic carcinoma. The result ofthis investigation can be the theoretic basis for the further step research,such as animal experiment and clinical experiment.
