Pancreatic cancer is one of the most malignant familiar alimentary tract cancer which have high malignant degree and early metastasis,incidence of a disease went on and on in the recent 20years.The key causation of the tumor' development is the growth of the tumor' blood vessel. While the growth of the tumor' blood vessel depend on regulation and domination of the so many vascular growth factors.Vascular endothelial growth factor(VEGF for short)is the most significant vascular growth factors whose coreceptor NRP-1 has been the hotspot of the study at present.We intend to discuss the expression and the correlation of NRP-1 and Micro vascular density in pancreatic cancer now.Objectives The aim of the study was to investigate the expression of the NRP-1 and to explore the possible correlation with Micro vascular density in pancreatic cancer.Methods We had selected embedded specimens from 40 patients who underwent operation for pancreatic cancer(exclude perampullary adenocarcinoma)in the first affiliated hospital of Shan Xi medical University and Shan Xi tumor hospital from 1999 March to 2005 October. Which included male were 22 cases and were female 18 cases.The location of the tumor were that cancer of head of the pancreas were 25 cases and cancer of body and cauda of tue pancreas were 15 cases.The pathologic type was that high was lcase,middle differentiation were 24 cases and low differentiation were 15 cases,while there were 15 casas had lymphatic metastasis.We would have to select another 25 specimens of natural pancreatic tissue to compare the pancreatic cancer mentioned above.Immunohistochemical method was used to detect all of the specimens'expression of the NRP-1 and Micro vascular density(MVD for short).The intension of the NRP-1'expression was detected by average ray density.The method of caculated MVD we would refer to the method which Weidner and Maeda had reported.Elected the location of the highest density of tumor blood vessel in 100×.Then elected brown colored endothelial cell or cell cluster which were different from other tumor cell and connective tissue obviously as one blood vseesl unit.We would caculate quantity of the blood vseesl unit in 5 fields of vision and make an average on that.Results(1)The positive rate of NRP-1 expression in pancreatic cancer was 92.5%(37/40) and that in normal or benign lesion pancreatic tissue was 8%(2/25).There was statistical difference between them(p<0.001).(2)The quantity of MVD in pancreatic cancer and normal or benign lesion pancreatic tissue were 46.70±6.454,23.08±4.26,respectively.There was statistical difference between them(p<0.05).(3)There was no significant difference among the expression of NRP-1 in different sex,ages,tumor location,tumor diameter,tumor sort of the cancer and lymphatic metastasis.However there was a relationship with histodifferentiation.(4)There was also no significant difference among the expression of MVD in different sex,ages,tumor location,tumor diameter,tumor sort of the cancer and and lymphatic metastasis.However there was a relationship with histodifferentiation.(5)There was significant positive correlation between NRP-1 and MVD expression(γ=0.878,P<0.001). (6)There was no positive correlation between NRP-1 and MVD expression(γ=0.332,P>0.05).Conclusions(1)NRP-1 was highly expressed in pancreatic cancer and the expression of NRP-1 was associated with histodifferentiation.(2)MVD was highly expressed in pancreatic cancer and the expression of MVD was also associated with histodifferentiation.(3)NRP-1 expression was positive correlated with MVD expression.
