ObjectiveStudy on the immunoregulation effect, acute toxicity and 30days sub acute toxicityof Ganoderma lucidum spores oil elastic capsule on normal mice, to elucidate themechanism action of Ganoderma lucidum spores oil elastic capsule in the immuno-regulation effect, understand of its safety, and provide the pharmacodynamic andtoxicology evidences of Ganoderma lucidum spores oil elastic capsule for newanti-cancer drug research and supplication in clinic.Methods1. The mice were divided into control (orally given maize oil) and 3 Ganodermalucidum spores oil groups (at low, moderate and high doses), orally given forconsecutive 30 days. The body, spleen, thymus weight were observed, study on theeffect of the drug on the cellular immune function by proliferation and transformationof spleen lymphocytes test; study on the effect of the drug on the humoral immunefunction of mice by serum erythrocytolysin test; study on the effect of the drug on themononuclear macrophage function by carbon clearance and neutral red test. Study onthe activity of NK cells, the level of TNF-α, IL-2 in spleen and the main organpathology by MTT reduction assay method.2. 20 mouse were divided into control and Ganoderma lucidum spores oil group, eachgroup have 5 female and 5 male, were orally given Normal saline and maximumvolume and concentration of Ganoderma lucidum spores oil. The acute toxicity andmain organs of pathological section for 30 days were observed.Result1. Ganoderma lucidum spores oil at 0.20g/kg, 0.40g/kg, 1.20g/kg dose couldsignificantly promote the lymphocyte transformation, raise clearance index numberand phagocytic index number, enhance the activity of NK cells and the level of TNF-α, IL-2 in spleen. Ganoderma lucidum spores oil at high dose and middle dose couldsignificantly enhance the serum erythrocytolysin level.2. There was no death and other abnormal reaction when the mice were orally giventhe maximum volume and concentration of Ganoderma lucidum spores oil in theacute toxicity test, which indicated its toxicity is small. Abnormality of main organ inpathological section for 30days sub acute toxicity was not been observed obviously.Conclusion1. Ganoderma lucidum spores oil can enhance the immune function of normal mice.2. Ganoderma lucidum spores oil was safe.
