Objective: To investigate the expression of PTEN,bFGF and microvessel density and their dependabilities and its clinical significance in prostatic carcinoma;to approach the contribution of Angiogenesis of PTEN and bFGF during the course of genesis and development of prostatic carcinoma.Methods: PTEN and bFGF protein expression and microvessel density in 23 normal prostate(NP),42 benign prostate hyperplasia(BPH) and 64 prostae carcinoma(Pca) was examined with immunohistochemistry.Results: (1) In NP and BPH groups,the total positive rate PTEN expression was 100%,while it expressed only in 28.13% of the prostate carcinoma which was significantly lower than the former two groups(P<0.01).The expression of bFGF was on the rise from NP to BPH and Pca(P<0.01), which was 0%, 59.70% and 76.56% respectively.(2) In Pca group, the expression of PTEN was 28.13%, which was closely related to the grade and clinical stage of prostate carcinoma.The higher the pathological grade and clinical stage of prostatic carcinoma were the lower expression of PTEN(P<0.01). There were no significant differences between highly and moderately cancer(P>0.05), while the differences between highly, moderately differentiated and poorly differentia- ted cancer were significant(P<0.05). The expression of bFGF was 76.56%, which was relevant to the clinical stage of prostatic carcinoma(P<0.05)but not to the grade (P>0.05).The expression of PTEN and bFGF was negatively correlated in prostate carcinoma(r=-0.802, P<0.01).(3) In NP group the average MVD was 16.90±3.69, and BPH group 27.54±2.24, Pca group 59.37±15.87.The MVD averages there were significant differences between NP group and BPH group(P<0.01),and the MVD averages of both NP group and BPH group there were significant differences with PCa group(P<0.01).The microvessel enumerations in prostatic carcinoma were increasing followed by the pathological grade and clinical stage of prostatic carcinoma increasing,that was negative related with PTEN expression(r=-0.681, P<0.01) and positive related with bFGF expression(r=0.708, P<0.01).(4) In the metastasized and non-metastasized lymphonodes PCa group the expressi- on of PTEN and bFGF was 35.29%,0%,70.59%,100.00% respectively.These were closely related to the lymphonode metastasis of prostate carcinoma (P<0.05).Conclusion: (1) Absence of PTEN expression is closely related to the initiation and proliferation of prostate carcinoma. PTEN expression is decreasing followed by the pathological grade and clinical stage of prostatic carcinoma increasing.And the detection of PTEN may contribute to the pathological grade,clinical stage and prognostic judgement of prostatic carcinoma.(2) bFGF is a phenotype of malignancy in prostate carcinoma that is closely related to the clinical staging and lymphonode metastasis, which suggests that high bFGF expression could promote the development of prostate carcinoma.(3)The expression of PTEN is negatively related with the expression of bFGF and MVD in prostatic carcinoma,which indicates that inactivation of PTEN can up-regulate bFGF expression and promote angiogenesis and development in prostatic carcinoma.
