ObjectiveThe expression of COX-2,bFGF was detected in the surgery gastric carcinoma paraffin tissues and normal carcinoma-adjacent tissues by using immunohistochemistrial SP method, which was to investigate the correlation among the expression,, the clinical pathologic characteristic and about angioge-nesis, and to provide the theory basis for the tumor antiangiogenesis treatment.Materials and methodsThe expression of COX-2, bFGF was detected in 38 cases of stomach carcinomatous tissues and 36 cases of normal carcinoma-adjacent tissues (which was over 5cm farer).All of the tissues choosing 4 pieces each slice were collected randomly from primary surgical procedures in our hospital (Tai'an city central hospital) from January 2009 to August 2009, which were confirmed by pathology and the data was intact, and the latter was looked as a control group.The CD34 monoclonal antibody was marked microvascular number of vascular endothelial cell count per unit area. The results were analysised by SAS 9.0, and p<0.05 was recognized as the criterion of statistical significance.Results(1) COX-2 was highly expressed in gastric carcinoma tissues with 71.05%, while it was expressed as 0 in carcinoma-adjacent tissues (which was over 5cm farer), not being detected the expression of COX-2 in them.The positive rate of COX-2 in stagesⅢ-Ⅳwas significantly higher than those inⅠ-Ⅱ(respectively p<0.01).There was a significant difference statistically (p<0.01). The positive rate of COX-2 in metastatic group was higher than that in non-metastatic group (p<0.05). The differences were significant statistically. The expression of COX-2 was not related to the patient's age, sex, tumor size, tumor location, differentiated degrees and the depth of invasion(p<0.05). (2)The positive rate of bFGF in gastric carcinoma tissues was 60.53%, while it was expressed with 27.78% in carcinoma-adjacent tissues.There was significant difference (p<0.05).The positive rate of bFGF in stagesⅠ-Ⅱin gastric carcinoma tissues was 22.22%, while the positive rate of bFGF in stagesⅢ-Ⅳin gastric carcinoma tissues was 72.41%.There was no difference statistically (p<0.01).The positive rate of bFGF in metastatic group(71.43%) was obviously higher than that in non-metastatic group, and there was a difference statistically (p<0.05).The expression of bFGF was not related to the patient's sex,age, tumor size, tumor location, differentiated degrees and the depth of invasion(p<0.05).(3)The result of microvessel density(MVD) marked with CD34 revealed that MVD(52.19±13.73)with lymph node metastasis in the tumor was higher than that (36.38±12.05)without lymph node metastasis;MVD(53.96±18.82) infiltrated to the deep group of carcinoma was higher than that (28.75±15.73)without infiltration to the deep one. Statistically,the difference was significiant(p<0.05).(4) The expression of COX-2, bFGF were correlated with MVD closely. MVD showed a rising trend with increasing expression.Conclusion1.COX-2 are over-expressed in gastric carcinoma,which is associated with the TNM classification and lymph node metastasis invasion and metastasis of gastric carcinoma, but they are not associated with age, sex,the depth of invasion and pathologic differentiation degree.2.The expression of bFGF in gastric carcinoma is significantly higher than those in normal mucosa, the expression of bFGF are associated with the TNM classification and lymph node metastasis,but they are not associated with age, sex, the depth of invasion and pathologic differentiation degree.3.It implys that there are probably synergistic effect on tumor angiogenesis to COX-2 and bFGF. Investigation through MVD along with COX-2, bFGF may help to determine classification and lymph node metastasis invasion of tumor.
