Objective To study the application of copper needle and explore the change of Cu2+ in serum and copper needles' effect to pathological and biochemical indicators of liver,kidney and heart before and after copper needles retained in centrial veins of rabbits' ears.So that it can be known more about whether the important organs of rabbits are effected by copper needles,and then better curing would be done in the future.Methods 49 rabbits were separated into 5 groups randomly,group A (which had been retained copper needles for 1d,n=10),group B(which had been retained copper needles for 3d,n=10),group C(which had been retained copper needles for 5d,n=10),group D(which had been retained copper needles for 7d,n=10)and control group(without being retained any copper needles, n=9).The rabbits of each group were arranged respectively to draw some vein blood to detect ion concentration of Cu2+([Cu2+])and biochemical indicators of heart creatase,liver and kidney function,which include AST,ALT,BUN, Cr,CK,CK-MB,LD and LD1.And then one group of rabbits were killed by injecting some air into their hearts directly,at the same time a piece of rabbits' livers,kidneys,heart and ear centrial veins were cut away as specimens.All of them were done some pathological examinations.Some samples with masculine expression of them also were observed after beening dealed through histological chemistry specific stained(Masson dyeing and Ag dyeing)and checked by transmission electron microscope.Results1 In contrast with the condition of ion concentration of Cu2+before copper needles beening retained in,it increased gradually when the time went. The ion concentration of Cu2+reached its maximum concentration when copper needles having been in centrial vein of rabbit's ears for 7 days (P<0.05).2 Compared with normal expression,it could been observed clearly and directly after copper needles beening retained that local rabbit ear tissue where copper needles were in became red and swollen,at last getting purple red.It could be found from specimens being HE dyed that wall of veins were destroyed and disappeared partly or completely and thrombus in veins came into being gradually.At the last of this period,short and small dot or bundle fibers were observed inside the veins with specific dyeing.3 There was not significant change of ALT and AST compared with before (P>0.05),but both of them shew fairly fort positive correlation on the first,fifth and seventh day after copper needles being retained(P<0.05); there was extinctive generally aseprtic inflammation in the part of liver, without construction of hepatic lobules changing and fibrous degeneration happening.4 There was significant change of BUN,one of kidney function parameter, compared with before(P<0.05).The result of BUN expressed temple increasing in seven days when copper needles were retained in the rabbits' ear central veins.However,there was not significant change of Cr,another important kidney function parameter,compared with before (P>0.05).Observed by light microscope,almost all of kidney specimens shew nothing with abnormalities,except very few of them.5 In contrast with before,all of them,CK,CK-MB,LD and LD1,changed significantly after copper needles being retained.Although fluctuated,all descended mainly.Observed by light microscope,almost all of heart specimens shew nothing with abnormalities,except very few of them.Conclusion1 Copper needles can release Cu2+gradually,which increased when the time went.2 During the experiment,the body response to copper needles mostly happens in the position where the copper needles being retained,hardly with little response to the whole body.But it could lead to slight, reversibility pathologic changes of liver and temple increasing of BUN, one of important kidney power parameter.
