The Study Of The Impact Of Different Lipid Lowering Scheme On The Liver And Kidney Function And Endothelial Function In Rats Arteriosclerosis Model

Objective：The purpose of this study is to observe the effect of endothelialfunction and the function of liver and kidney protective,when use single largedose of simvastatin and small doses of simvastatin combined with ezetimibe totreat the mixed hyperlipidemia rats.Method：A number of44health6months male Wistar rats are dividedinto five groups at random：normal control group、high lipid control group、Simvastatin (10mg·kg^-1·d^-1) treatment group、Simvastatin (20mg·kg^-1·d^-1)treatment group and Simvastatin (10mg·kg^-1·d^-1) joint Ezetimibe(5mg·kg^-1·d^-1)treatment group.Then the rats are given corresponding doses of simvastatin andezetimibe by irrigating stomach. Finally,determinate the level of TC、LDL-C、HDL-C、TG、ALT、AST、Scr、BUN、UA、U-pro、Ucr、CCr、CRP、sTM、sICAM-1and urinary protein concentration.The aortic and renal tissues of Ratswere stained with HE staining and calculated renal index.Result：1.Each treatment group can reduce the level of TC、LDL-C、TG、ALT、AST、BUN、UA、U-pro、CRP、sTM and sICAM-1,while raise the levelof HDL-C and Ccr significantly than high lipid treatment group. Simvastatin(20mg·kg^-1·d^-1) treatment group can better improve the renal function andreduce the level of sTM and sICAM-1.2.Optical observations hints, Thetreatment groups is smoother than model group. glomerular volume of thetreatment groups are smaller than high lipid control group, and the proliferationof mesangial cell are reduced.At the same time, the degeneration and necrosisof renal tubular epithelial cell and extracellular matrix components are reduced too.Simvastatin (20mg·kg^-1·d^-1) treatment group improved more significantly.Conclusions：1.Single large dose of simvastatin compare with small doses ofsimvastatin joint ezetimibe can better reduce the level of BUN, UA, U-pro,sTM and sICAM-1, increase the level of Ccr and HDL-C, and can betterimprove glomerular morphology,and improve endothelial function.2.Small dose of simvastatin joint ezetimibe compare with a single largedose of simvastatin can significantly reduce the level of TC, LDL-C and CRPsignificantly without any increasing in serum transaminase levels, does notincrease the damage of liver function.
