PART 1 The empirical study of the activity of NF-κB and the expression of TNF-αand IL-1βin rat CNPObjection: To investigate if the activity of NF-κB and the expression of TNF-αand IL-1βincrease in CNP and approach their role in the onset of CNP.Method: 20 1-year-old male Sprague-Dawley rats were divided randomly into control group and model group. The model group had been castrated and injected with estradiol benzoate (0.25 mg/kg/day, s.c.) for 30 days, the control group were treated with sham operation and then placebo. After the modeling had been done,the prostates of the two groups were taken. The Pathologic appearance of the prostate tissue of the two groups were observed with HE staining and light microscope.And the location and the positive cell rate of NF-κB and the expression of TNF-αand IL-1βof the two groups were detected with immunohistochemical method.Results: The control group did not have inflammatory reaction in their prostates,while the model group displayed visible inflammatory reaction. NF-ΚB located in cytoplasm predominantly in the control group, while translocated from cytoplasm to nucleus obviously in the model group. And the positive cell rate of NF-κB also increased obviously in the model group. There was significant difference of the positive cell rate of NF-κB between the model group and the control group(P<0.01).the expression of both TNF-αand IL-1βwas negative in the control group,while strongly positive in the model group. There was significant difference of the expression of both TNF-αand IL-1βbetween the model group and the control group,according to the result of photodensitometry and area density analysis(P<0.01).Conclusion: The activity of NF-κB and the expression of TNF-αand IL-1βincreased in CNP. overactivation of NF-κB and overexpression of TNF-αand IL-1βmay result in focal immune imbalance and Inflammatory reaction in prostate tissue and play a important role in the onset of CNP.PART 2 The empirical study of the therapeutical effect and mechanism of action of NAC in CNPObjection: To investigate the therapeutical effect and the mechanism of action of NAC in CNP and provid a new strategy for clinic management.Method: 30 1-year-old male Sprague-Dawley rats were divided randomly into control group , model group and intervention group. The model group and the intervention group had been castrated and injected with estradiol benzoate (0.25 mg/kg/day, s.c.) for 30 days,and the control group were operated shamly and injected with physiologic saline.After the modeling had been done , the intervention group were administrated intragastricly with NAC(125mg/kg/day) for 7 days,and the model group and the control group with physiologic saline.Then the prostates of the three groups were taken.The Pathologic appearance of the prostate tissue of the three groups were observed with HE staining and light microscope.And the location and the positive cell rate of NF-κB and the expression of TNF-αand IL-1βof the three groups were detected with immunohistochemical method.Results: The control group did not have inflammatory reaction in their prostates.the model group displayed visible inflammatory reaction. Inflammatory reaction remitted obviously in the intervention group. The activity of SOD ( P<0.05)decreased and the contents of MDA( P<0.01)increased in the model group, compared with the control group. The activity of SOD ( P<0.05)increased and the contents of MDA (P<0.01)decreased in the intervention group, compared with the model group. But significant difference of both the activity of SOD and the contents of MDA was not shown between the intervention group and the control group ( P>0.05 ) . NF-ΚB located in cytoplasm predominantly in the control group. In the model group NF-ΚB translocated from cytoplasm to nucleus obviously. In the intervention group nucleus staining weakened. The positive cell rate of NF-κB : the model group> the intervention group> the control group (P<0.01). The expression of TNF-αand IL-1β: the model group> the intervention group> the control group (P<0.01).Conclusion: the activity of SOD lowered , the contents of MDA rised and oxidative stress accordingly existed in prostate tissue in CNP. NAC could raise the activity of SOD , lower the contents of MDA, relieve the oxidative stress in prostate tissue in CNP, thus down-regulated the activity of NF-ΚB and lowered the expression of TNF-αand IL-1βand resulted in remission of inflammatory reaction and displayed therapeutical effect on CNP.
