Experimental Study Of The Nerve Growth Factor NGF And Inflammatory Factor TNF-α On Chronic Nonbacterial Prostatitis Treated By Qian Mi Tong

Objective:To evaluate the therapeutic effect of the experimental study that rats on chronic nonbacterial prostatitis treated by Qian Mi Tong, to investigate the effect of the nerve growth factor NGF, inflammatory factor TNF-a and organize pathological changes on chronic non bacterial prostatitis that treated by Qian Mi Tong and to provide experimental and theoretical basis for the development and application of traditional Chinese medicine in the treatment of chronic non bacterial prostatitis.Method:60 male rats,10-12 months old and Weight 100-150g, domesticating and feeding a week, were randomly divided to 6 groups, each of 10 (blank group, model group, positive control group, Qian Mi Tong low/medium/high dose group).Except the blank group, the rest of the group rats was given with intraperitoneal injection of anesthesia and castration operation under sterile conditions, continued to give subcutaneous injection of estradiol benzoate 0.25mg/kg the day after the operation, once a day for 30 days.Last time of subcutaneous injection of estradiol benzoate ended and gavage began next day, blank group and model group were intragastrically infused with a saline content equaled to the Chinese Medicine content of experiment group; control group:each rat was given by intragastric administration of Qian Lie Tong Yu capsule suspension (3.8g/kgd) equaled to 20 times of the amount of the adult weight 70Kg; Qian Mi Tong low/medium/high dose group:each rat was given by intragastric administration of Qian Mi Tong suspension (1.9g/kg·d、3.8g/kg·d、7.6g/kg·d) equaled to 5 times,10 times,20 times of the amount of the adult weight 70Kg.All groups were intragastrical orally once for 30 days.24 hours after the last administration, said the weight of rat, than put it to death and removed the prostate tissue, said its wet weight and calculate the prostatic index.Fixed the prostate tissue by 4% formalin, made slices and dyed, observed the pathological changes in prostatic tissue under the light microscope and used the immunohistochemistry to detect the expression of NGF and TNF-a protein in prostatic tissue.ResultrCompared with the blank group, the body weight of rats in drug groups significantly decreased (P<0.01); compared with the model group, the prostate wet weight and the prostate index of drug groups rats were significantly decreased (P<0.01), the NGF protein expression(P<0.01) and the TNF-a protein expression(P<0.05) in prostate tissue which was intragastrically infused with high/medium dose group of Qian Mi Tong significantly reduced, pathological changes in prostate tissue which was intragastrically infused with high/medium dose group of Qian Mi Tong significantly reduced.ConcIusion:Qian Mi Tong has obvious therapeutic effect on chronic nonbacterial prostatitis rat, its mechanism is related with the reduction of NGF、TNF-a and pathological changes in prostate tissue of chronic nonbacterial prostatitis rat.
