[Objective]: To study the expression of KAI1 in human gastric carcinoma, explore the relationship between KAI1 expression and clinicopathological features in human gastric carcinoma;to study the expression of PTEN and CD44V6 in human gastric carcinoma, explore the relationship between PTEN and CD44V6 expression and clinicopathological features in human gastric carcinoma;and to find out the relationship between KAI1,PTEN and CD44V6 in gastric carcinoma..[Method]:1.The expression of KAI1mRNA in human gastric carcinoma tissues and normal gastric tissues was detected by reverse transcriptase polymerase chain reaction(RT-PCR).2.The expression of KAI1,PTEN and CD44V6 in human gastric carcinoma tissues and normal gastric tissues were detected by S-P immunohistochemistry.[Results]:1.The expression rate of KAI1mRNA in 61 cases of gastric carcinoma and normal gastric tissues were 80.3%(49) and 85.2%(52), no statistical difference was found between them (P>0.05);their expression levels of KAI1mRNA were 0.470±0.239 and 0.527±0.225,no statistical difference was found between them either(P>0.05).2.The expression rate of KAI1 protein was significant lower in gastric carcinoma tissues(29.5%,18/61) than normal gastric tissues(90.0%,18/20),statistical difference was found between them (P<0.01).The expression rate of PTEN protein was significant lower in gastric carcinoma tissues(45.9%,28/61) than normal gastric tissues(95.0%,19/20),statistical difference was found between them (P<0.01).The expression rate of CD44V6 was significant higher in gastric carcinoma tissues(65.6%,40/61) than normal gastric tissues(5.0%,1/20),statistical difference was found between them (P<0.01). 3.The expression of KAI1 protein,PTEN protein and CD44V6 were associated with the depth of tumor invasion,differentiation grade,lymph node metastasis and TNM stage (P<0.05) but had no correlation with age and sex(P>0.05).4.There were 17 cases(34.7%) of KAI1 protein positive expression in 49 cases of KAI1mRNA positive expression,while there were 32 cases(65.3%) of KAI1 protein negative expression in 49 cases of KAI1mRNA positive expression.No correlation was found between the expression of KAI1mRNA and KAI1 protein(P>0.05).5.The KAI1 protein shew significant positive correlation with the PTEN protein expression in gastric carcinoma (r=0.342,P<0.01). The KAI1 protein shew significant negative correlation with the CD44V6 expression in gastric carcinoma (r=-0.363,P<0.01).No correlation was found between the expression of PTEN and CD44V6 in gastric carcinoma (P>0.05).[Conclusion]:1. The expression of KAI1mRNA in gastric carcinoma tissues and normal gastric tissues had no statistical difference; The expression of KAI1mRNA had no correlation with the age,sex,depth of tumor invasion,differentiation grade,lymph node metastasis and TNM stage.That suggest the carcinogenesis,progression,invasion and metastasis of gastric carcinoma had no correlation with the KAI1mRNA expression.2. The expression of KAI1 protein and PTEN protein were significant lower in gastric carcinoma than normal gastric tissues. The expression rate of CD44V6 was significant higher in gastric carcinoma than normal gastric tissues. The expression of KAI1,PTEN,CD44V6 were associated with the depth of tumor invasion , differentiation grades, lymph node metastasis and TNM stage.That shew KAI1 protein and PTEN protein suppressed the processes of carcinogenesis,progression,invasion and metastasis of gastric carcinoma,while CD44V6 promoted it. Downregulation of KAI1 protein,PTEN protein and upregulation of CD44V6 promoted the processes of carcinogenesis,progression,invasion and metastasis of gastric carcinoma,Which made the gastric carcinoma cells more malignant and more invasive and easier to metastasis. 3.The downregulation of KAI1 protein in the gastric carcinoma tissues had no correlation with KAI1mRNA. The downregulation of KAI1 protein in the gastric carcinoma tissues had no correlation with the gene transcription and mutation ,but may had correlation with the mRNA translational dysregulation or posttranslational modification dysregulation.4. KAI1 protein and PTEN protein interpromoted in the processes of carcinogenesis,progression,invasion and metastasis of gastric carcinoma,while,KAI1 protein and CD44V6 interrestricted in it.5.KAI1 protein,PTEN protein and CD44V6 can be the indicators to judge the biological behavior of gastric carcinoma.
