PART ONE Investigation of BDV Natural Infection in rabbits in ChongqingObjective:To investigate the recessive infection of Borna Disease Virus in rabbits in chongqing, analyze their origin,and explore if the nucleotide sequence and amino acid sequence in BDV P24 were homophylic with overseas strains, including standard strain V/FR , strain He/80/FR and strain H1766.Methods: BDV P24 fragment was detected by nested reverse transcriptase polymerase chain reaction with real-time PCR(Real-time nRT-PCR) in brain tissues from 60 rabbits. For those positive products, the gene sequence and amino acid sequence were then analyzed by BLAST and DNAstar V6.13 EditSeq.Results: The positive rate was 3.33% (2/60). The gene sequence for positive products showed BDV P24 in rabbits from Chongqing was homophylic with strain H1766 detected from ill horses (97.67%) and also with the standard strain V/FR and strain He/80/FR (96.51%), however, their amino acid sequence remained the same.Conclusion: There is BDV recessive infection, which probably originate from animals, in rabbits of Chongqing. The gene sequence is homophylic with that of standard strain V/FR and strain He/80/FR especially with strain H1766.PART TWO Investigation of BDV Natural Infection in dogs in ChongqingObjective:To investigate the recessive infection of BDV in dog in Chongqing , analyze their origin,and explore if the nucleotide sequence in BDV p24 was homophylic with the overseas strains, including standard strain V/FR, strain He/80/FR and strain H1766, detected from human and animals.Methods: The p24 fragment of BDV was detected by Real-time nRT-PCR in brain tissue (right prefrontal lobe) from 40 dogs. And then the products of Real-time nRT-PCR from positive samples were cloned and sequenced before compared with those of the overseas strains, including strain V/FR, strain He/80/FR,strain H1766 by BLAST and DNAstar V6.13 EditSeq.Results: There are 5 positive samples in 40 dogs. The gene sequence for positive product showed BDV p24 in dogs from Chongqing was homophylic with strain V/FR and strain H1766 detected from ill horses (97.67%) and strain He/80/FR (94.19%) . Interestingly , it is only 95.35% homology with P protein gene isolated from Austrian dogs. however, their amino acid sequence remained the same.Conclusion: There is BDV recessive infection, which probably originate from animals, in dogs of Chongqing. The gene sequence is homophylic with that of standard strain V/FR , strain H1766 and strain He/80/FR .PART THREE Investigation of BDV Natural Infection in pigeon in ChongqingObjective:To investigate the recessive infection of BDV in pigeon in Chongqing , analyze their origin,and explore if the nucleotide sequence in BDV p24 was homophylic with the overseas strains, including standard strain V/FR, strain He/80/FR and strain H1766, detected from human and animals. Methods: The p24 fragment of BDV in PBMCs from 60 pigeons was detected by Real-time nRT-PCR. And then the products of Real-time from positive samples were cloned and sequenced before compared with those of the overseas strains, including strain V/FR, strain He/80/FR,strain H1766 by BLAST and DNAstar V6.13 EditSeq.Results: There is 4 positive samples in 60 pigeons. The gene sequence for positive products showed BDV p24 in pigeon from Chongqing was homophylic with strain He/80/FR(96.51%) , strain V/FR(95.35%)and strain H1766 (95.35%). however, their amino acid sequence remained the same.Conclusion: There is BDV recessive infection, which probably originate from animals, in pigeon in Chongqing. The gene sequence is homophylic with that of standard strain V/FR , strain He/80/FR and strain H1766.
