OBJECTIVE: To investigate the expression and significance of E-cadherin, VEGF and CD44V6 in colorectal carcinoma.METHOD: The expression of E-cadherin,VEGF and CD44V6 among colorectal carcinoma,colonic adenoma and normal colonic tissue were tested by immunohistochemical assay.RESULT: The positive rate of E-cadherin in colonic cancer was 53.2%, the expression was lower than colonic adenoma and normal colon tissue (P<0.05). In colorectal carcinoma, the expression of CD44V6 and VEGF was higher than colonic adenoma and normal colon tissue (P<0.05), the positive rate of CD44V6 and VEGF were 41.9% and 58.1%, respectively. The expression of E-cadherin in colonrectal carcinoma was correlated with lymph node metastasis , the Duke's clinical stage (P<0.05), and not associated with the degree of tumor differentiation, the depth on invasion, patient's age and tumor locus(P>0.05). The expression of CD44V6 in colorectal carcinoma was correlated significantly with the lymph node metastasis, the Duke's clinical stage and the depth on invasion (P<0.05), but it was not associated with the degree of tumor differentiation, patient's age and tumor locus (P>0.05). The expression of VEGF in colon cancer was correlated significantly with the lymph node metastasis, the Duke's clinical stage, the depth on invasion and the degree of tumor differentiation (P<0.05), but it was not associated with patient's age and tumor locus(P>0.05). There was no relationship between the positive rate of E-cadherin and CD44V6. A positive correlation was found between CD44V6 and VEGF expression (r=0.457, P<0.01), while a negative correlation was found between E-cadherin and the VEGF expression (r=-0.60, P<0.01).CONCLUSION: Low expression of E-cadherin and Overexpression of CD44V6 and VEGF play an important role in the process of invasion and metastasis of colorectal cancer. These data show that E-cadherin is a tumor non-metastatic gene, while VEGF and CD44V6 are the tumor trigger metastasis gene.
